The orange-spotted grouper () is an important mariculture fish in China. However, in recent years, with the rapid development of aquaculture activities, outbreaks of viral diseases have affected the grouper aquaculture industry, causing severe economic losses. In the present study, we isolated and identified a virus from diseased, orange-spotted groupers from an aquaculture farm in Hainan Province, China.

Article Synopsis
  • Tick-borne encephalitis (TBE) outbreaks in Europe have been linked to the consumption of raw milk products, highlighting a need for better detection methods of the virus in these foods.
  • Researchers developed a sensitive cell culture-based method for detecting infectious TBEV in artificially contaminated raw goat milk and cheese, achieving varying levels of viral detection.
  • The study showed that this method successfully identified infectious TBEV in both milk and cheese samples, supporting its potential use in future foodborne outbreak investigations.

Development of a Cell-Based Reporter Potency Assay for Live Virus Vaccines.

Vaccines (Basel)

July 2024

Analytical Research & Development, Merck & Co., Inc., Rahway, NJ 07065, USA.

The rapid development of potency assays is critical in the development of life-saving vaccines. The traditional plaque assay or fifty percent tissue culture infectious dose (TCID) assay used to measure the potency of live virus vaccines is time consuming, labor intensive, low throughput and with high variability. Described here is the development and qualification of a cell-based reporter potency assay for two vaccines for respiratory viral infection, one based on the recombinant vesicular stomatitis virus (rVSV) backbone, termed Vaccine 1 in this paper, and the other based on the measles virus vector, termed Vaccine 2.

Recently, a multiplex PCR-based titration (MPBT) assay was developed for simultaneous determination of infectious titers of all three Sabin strains of the oral poliovirus vaccine (OPV) to replace the conventional CCID assay, which is both time-consuming and laborious. The MPBT assay was shown to be reproducible, robust and sensitive. The conventional and MPBT assays showed similar results and sensitivity.

Immunotherapy has improved survival outcomes of patients with advanced melanoma. Lower gastrointestinal tract immune-related adverse events (irAEs) are common during treatment; however, gastritis is not frequently observed. Herein, we report a case of severe cytomegalovirus (CMV)-related gastritis in a patient treated with ipilimumab and nivolumab for metastatic melanoma.
