3D printed reservoir-like vaginal rings for antibiotic delivery.

Int J Pharm

January 2025

Sezione di Tecnologia e Legislazione Farmaceutiche "Maria Edvige Sangalli", Dipartimento di Scienze Farmaceutiche, Università degli Studi di Milano, via Giuseppe Colombo 71, 20133, Milano, Italy.

Targeting the development of a 3D printed reservoir-like vaginal rings (VRs) intended to fulfill the needs of precision medicine, prototypes ensuring prolonged release of metronidazole (MTZ) were preliminary manufactured and tested. Indeed, this drug represents the first-line therapy against bacterial vaginosis, which would especially benefit from convenient as well as easy dose adjustment and from more than 48 h continuous release, thus avoiding barely tolerated and repeated administrations. Starting from a soft thermoplastic elastomer (TPE), hollow ring structures were successfully printed at 190 °C and then extemporaneously filled with drug-loaded, in-situ-crosslinking hydrogel formulations based on alginate (ALG).

An Unexpected Case of Generalized Tetanus.

Cureus

December 2024

Intensive Care Unit, Unidade Local de Saúde da Região de Aveiro, Aveiro, PRT.

Tetanus is a disease of the nervous system caused by a toxin produced by , an anaerobe found in high concentrations in the soil. The occurrence of tetanus is related to contaminated traumatic wounds, and most patients have had some failure in their immunization. However, there are rare case reports of generalized tetanus in patients with proper vaccination schemes who failed to receive appropriate prophylaxis after high-risk exposure.

Cellulose Acetate Butyrate-Based In Situ Gel Comprising Doxycycline Hyclate and Metronidazole.

Polymers (Basel)

December 2024

Program of Pharmaceutical Engineering, Department of Industrial Pharmacy, Faculty of Pharmacy, Silpakorn University, Nakhon Pathom 73000, Thailand.

Cellulose acetate butyrate is a biodegradable cellulose ester bioplastic produced from plentiful natural plant-based resources. Solvent-exchange-induced in situ gels are particularly promising for periodontitis therapy, as this dosage form allows for the direct delivery of high concentrations of antimicrobial agents to the localized periodontal pocket. This study developed an in situ gel for periodontitis treatment, incorporating a combination of metronidazole and doxycycline hyclate, with cellulose acetate butyrate serving as the matrix-forming agent.

: The high recurrence rate of bacterial vaginosis (BV) after antibiotic treatment is at least partially attributed to resistant bacteria. The CAPRISA 083 (CAP083) study investigated the influence of metronidazole (MTZ) treatment on the vaginal microbiome in 56 South African women diagnosed with BV. To explore the etiology of recurrent BV in this cohort, we retrospectively analyzed vaginal swabs collected in CAP083 before and after MTZ treatment.
