Indian J Otolaryngol Head Neck Surg

January 2025

Department of Radiodiagnosis, IGMC, Shimla, 171001 Himachal Pradesh India.

Ewing's sarcoma is a rare and highly malignant tumour predominantly affecting the bones. Extra skeletal Ewing's sarcoma of Sino nasal tract, encompassing the nasal cavity and paranasal sinuses is an exceedingly rare location for this tumour. We present a case of a 16 year-old boy who presented with right nasal obstruction, epistaxis and right facial swelling since 3 months.

Background: Prior research has demonstrated that proteins play a significant role in the prognosis and treatments of various sarcomas, including Ewing sarcoma through the interplay of downstream signaling cascades. However, there is limited understanding about the strcucture conformation of EWSR1 and its structural implication in the prognosis of Ews-ing Sarcoma by interaction with RNA molecules.

Aims: The primary goal of ongoing research is to determine how EWSR1 contributes to Ewing sarcoma.

Although primary tumor control rates after surgery and/or radiation therapy (RT) are generally high in patients with Ewing sarcoma (EWS), those with unresectable tumors have failure rates approaching 30% and experience poorer outcomes. Additionally, although metastatic site irradiation is associated with improved survival, dose, and volume effects influence the long-term toxicity risk. Consequently, it is important to identify novel systemic agents to enhance the therapeutic ratio of RT.

Single-cell RNA sequencing provides a deep understanding of gene expression patterns at the single-cell level, enabling researchers to study cellular heterogeneity and differentiation. Spatial transcriptomics, meanwhile, delivers spatial visualization and quantitative analysis of tissue sections. Here, we introduce a protocol to isolate single cells and obtain high-quality spatial transcriptomics data from bladder Ewing sarcoma tissue.
