Ergothioneine (EGT) is a thiol-based antioxidant synthesized by certain fungal and bacterial species that is prevalent in the human diet. Recently, an EGT-specific transporter, EgtUV, was discovered in bacteria that are incapable of EGT biosynthesis, including the gastric pathogen Helicobacter pylori. However, EGT is naturally abundant in the complex media required to culture H. pylori and many other host-associated microbes, complicating efforts to understand how this molecule influences microbial physiology. Using the solute-binding domain of H. pylori EgtUV, we generated an EGT-chelating resin that depletes EGT from nutrient-rich media. We determined that wild-type H. pylori requires EGT to outcompete a transporter-deficient strain in vitro. Furthermore, EGT induces transcription of genes encoding outer-membrane transporters that may regulate intracellular EGT content upstream of the inner-membrane-localized EgtUV transporter. Our work establishes a method for tuning exposure to an abundant antioxidant in vitro, enabling future studies of EGT in diverse microbial strains and communities.

Introduction: The underlying cause of psoriasis, a chronic inflammatory skin condition driven by an immune response, remains a topic of active investigation and is not yet fully elucidated. Recent studies have revealed that ergothioneine, a small molecule sulfur-containing histidine derivative that can be ingested from the daily diet and accumulated in the body, exhibits antioxidant capacity that is comparable to that of glutathione. Nevertheless, there is a paucity of empirical data concerning the precise impact of ergothioneine in the context of anti-inflammatory processes, particularly in the context of psoriasis.

Ergothioneine (EGT), a naturally occurring histidine derivative, has been reported to modulate neurodegenerative diseases; however, the underlying mechanism remains unclear. This study aimed to investigate the brain-beneficial role of the natural amino acid EGT in NE-4C nerve cells. In the nerve cells, EGT treatment of >10 μM for 48 h significantly increased the expression of brain-derived neurotrophic factor (BDNF), as well as the phosphorylation of cAMP response element-binding protein (CREB), whereas no change was observed in acetylcholine receptor expression.

Background: The insertion of an enterogastric tube (oral or nasal) (EGT) is the passage of a tube through the nose or mouth into the stomach. In a paediatric setting, EGTs are used within clinical practice for a variety of reasons including enteral feeding, decompression, post-gastrointestinal surgery, patient assessment, and drug and fluid administration. Confirmation of EGT placement is required immediately following insertion and thereafter prior to each use, including after the administration of enteral feed or medication.
