The fruit of L. (Piperaceae), commonly used as a spice in various culinary applications, is renowned for its rich nutritional profile and potential medicinal properties. In this study, a molecular network-based dereplication strategy was employed to isolate 12 dimeric amide alkaloid enantiomers (-) from fruits, including eight pairs of cyclobutane-type dimers (-) and four pairs of cyclohexene-type dimers (-). Notably, five pairs of cyclobutane-type dimers, namely, piperlongramides E-I (-), and one pair of cyclohexene-type dimers, piperlongramide J (), were identified as undescribed compounds. The structures of these compounds were elucidated by comprehensive spectroscopic data, electronic circular dichroism (ECD) calculations, and X-ray diffraction analysis. Additionally, these compounds, for the first time, were subjected to chiral resolution. bioactivity screening revealed that compounds (+)-, (-)-, and (+)- exhibited notable anti-inflammatory effects in an LPS-induced RAW 264.7 macrophage model, with IC values of 23.42 ± 1.04, 32.72 ± 0.54, and 33.52 ± 1.75 μM, respectively. Furthermore, compound (+)- also demonstrated significant inhibitory activity againstα-glucosidase, with an IC of 11.69 ± 0.91 μM. Compounds (+)-, (+)-, and (-)-, exhibited promising inhibitory activity against PTP1B with IC values of 3.30 ± 0.09, 4.30 ± 0.18, and 4.37 ± 0.37 μM, respectively, indicating their potential antidiabetic effects. This study highlights the promising application of these amide alkaloid dimers in the development of functional foods and pharmaceutical products, thereby expanding the health-promoting potential of fruits.

Capsaicin, known for its antioxidant and antibacterial properties, may mitigate oxidative stress and inflammation associated with exercise-induced muscle damage (EIMD). This study evaluates the efficacy of capsaicin supplementation in reducing delayed-onset muscle soreness (DOMS) and enhancing strength and power in collegiate male futsal players. A randomized, double-blind, placebo-controlled crossover design was used.

Opioids are the most effective option for severe pain. However, it is well documented that the side effects associated with prolonged opioid use significantly constrain dosage in the clinical setting. Recently, researchers have concentrated on the development of biased opioid receptor agonists that preferentially activate the G protein signaling pathway over β-arrestin signaling.

Background: Viral myocarditis (VMC) is a cardiac condition characterized by inflammation of the myocardium due to viral infection. It is a significant cause of sudden cardiac death in young adults, and effective treatments remain limited. Inflammatory caspase-mediated pyroptosis serves as a host defense mechanism against pathogens and is crucial in the pathogenesis of VMC.

Introduction: It is common for finger pain in hand osteoarthritis (HOA) to display a neuropathic component. Non-steroidal anti-inflammatory drugs (NSAIDs) and conventional analgesics are not very effective in relieving this neuropathic-like pain. Capsaicin, a compound extracted from chilli peppers, is approved for the management of localised neuropathic pain.
