Bidirectional microscopy (BDM) combines simultaneous targeted optical perturbation and imaging of biophysical or biochemical signals (e.g. membrane voltage, Ca , or signaling molecules). A core challenge in BDM is precise spatial and temporal alignment of stimulation, imaging, and other experimental parameters. Here we present Luminos, an open-source MATLAB library for modular and precisely synchronized control of BDM experiments. The system supports hardware-triggered synchronization across stimulation, recording, and imaging channels with microsecond accuracy. Source code and documentation for Luminos are available online at https://www.luminosmicroscopy.com and https://github.com/adamcohenlab/luminos-microscopy . This library will facilitate development of bidirectional microscopy methods across the biological sciences.

The SARS-CoV-2 spike protein is synthesized in the endoplasmic reticulum of host cells, from where it undergoes export to the Golgi and the plasma membrane or retrieval from the Golgi to the endoplasmic reticulum. Elucidating the fundamental principles of this bidirectional secretion are pivotal to understanding virus assembly and designing the next generation of spike genetic vaccine with enhanced export properties. However, the widely used strategy of C-terminal affinity tagging of the spike cytosolic tail interferes with proper bidirectional trafficking.

The intraflagellar transport (IFT) machinery, containing the IFT-A and IFT-B complexes and powered by dynein-2 and kinesin-2 motors, is crucial for bidirectional trafficking of ciliary proteins and their import/export across the transition zone (TZ). Stepwise assembly of anterograde IFT trains was proposed previously; that is, the IFT-B complex first forms a TZ-tethered scaffold with sequential incorporation of IFT-A, dynein-2, and finally kinesin-2. However, IFT-A and IFT-B complexes also demonstrate distinct localization to the basal body/mother centriole.

Colon cancer is a prevalent malignancy, substantially it prevented most effectively from killing patients through early endoscopic detection. With the rapid development of artificial intelligence technology, the early diagnosis rate of colonic polyps achieves greater clinical efficacy for colon cancer by applying target detection algorithms to colonoscopy images. This paper presents two outcomes achieved through the application of the improved YOLOv5s algorithm with annotated microscopy images of clinical cases and publicly available polyp image data: (1) enhancement of the C3(Cross Stage Partial Networks) module with multiple layers to C3SE(Cross Stage Partial Networks with Squeeze-and-Excitation) via the attention mechanism SE (squeeze-and-excitation) and (2) fusion of higher-level features utilizing BiFPN (the weighted bi-directional feature pyramid network).
