In vitro inhibitory activity of indole alkaloid derivatives against porcine epidemic diarrhea virus.

Porcine epidemic diarrhea virus (PEDV) is an enteric coronavirus that can cause acute diarrhea, vomiting, dehydration, and high mortality of newborn piglets, leading to huge economic losses to the world pig industry. Given the limited efficacy of current PEDV vaccines, there is an urgent need for the development of antiviral drugs. In this study, the antiviral effects of 17 synthesized indole alkaloid derivatives against PEDV were investigated. It was observed that indole alkaloid derivative no. 14 exhibited significant inhibition of PEDV replication in a dose-dependent manner. Furthermore, time-of-addition assays and quantitative real-time PCR (QPCR) showed that delayed administration of this compound resulted in a weaker inhibitory effect on PEDV compared to early treatment. Mechanistic analysis revealed that this compound exerts its inhibitory effects during the entry stage of the PEDV life cycle. This study demonstrates the anti-PEDV effects of indole alkaloid derivative no. 14, suggesting its potential as a candidate drug for treating PEDV infections.

This study investigates the metabolic responses of cancerous (RCC) and non-cancerous (HK2) kidney cells to treatment with Staurosporine (STAU), which has a pro-apoptotic effect, and Bongkrekic acid (BKA), which has an anti-apoptotic effect, individually and in combination, using H NMR metabolomics to identify metabolite markers linked to mitochondrial apoptotic pathways. BKA had minimal metabolic effects in RCC cells, suggesting its role in preserving mitochondrial function without significantly altering metabolic pathways. In contrast, STAU induced substantial metabolic reprogramming in RCC cells, disrupting energy production, redox balance, and biosynthesis, thereby triggering apoptotic pathways.

The identification of these classes is of particular relevance to drug discovery, as they could serve as valuable starting points in developing effective and safe anti-inflammatory agents. The privileged compound classes of natural products include the polyphenols, coumarins, labdane diterpenoids, sesquiterpene lactones, isoquinoline and indole alkaloids, each offering a variety of molecular scaffolds and functional groups that enable diverse interactions with biological targets.

Objective: To study the changes in positron emission tomography with 2-deoxy-2-[fluorine-18]fluoro-D-glucose integrated with computed tomography (F-FDG PET/CT) aortic target-to-background ratio (TBR) and aortic calcification scores before and after 6 cycles of chemotherapy with the rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone (R-CHOP) regimen in patients with diffuse large B-cell lymphoma (DLBCL).

Patients And Methods: We selected 161 patients with DLBCL who received 6 cycles of R-CHOP standard chemotherapy and underwent baseline and 6-cycle efficacy evaluations using F-FDG PET/CT examinations at the Second Hospital of Dalian Medical University from July 2017 to June 2023. Additionally, 125 patients who underwent F-FDG PET/CT for physical examination during the same period, without active malignancy or systemic inflammatory disease, were chosen as the control group.
