Background: Sperm motility and chemotaxis are important early steps in the interaction between sperm and oocytes during fertilization. Understanding these processes is essential for their basic biological and clinical applications. This review outlines advances in understanding the molecular mechanisms of sperm activation and chemotaxis over the past two decades.

Methods: The review focuses on the molecular pathways of Ca signaling and the role of the CatSper channel involved in this signaling, and examines the comprehensive mechanisms that regulate sperm motility in aquatic invertebrates, fish, and mammals.

Main Findings: Sperm are activated by environmental changes (e.g., pH and osmolality) and egg-derived factors. CatSper channels mediate Ca influx and regulate cell motility and chemotaxis. In addition to Ca, cAMP and membrane potential are also involved in the regulation of sperm motility. Alternative pathways exist in species lacking CatSper, highlighting the diversity of sperm activation mechanisms.

Conclusion: There has been significant progress in understanding sperm motility regulation mediated by Ca, notably with CatSper, but the molecular mechanisms of other factors remain unclear. Future research should focus on species lacking CatSper to uncover commonalities and diversity in sperm motility regulation using genome editing and transcriptomic analyses.

Yak is a large plateau mammal with low reproduction rate, while cryptorchidism is a major reproductive disorder associated with infertility in highland yaks. To better understand the occurrence of cryptorchidism and its regulatory mechanisms in yaks, we conducted a multi-omic analysis, and screened a total of 4456 differentially expressed genes (DEGs) and 332 differentially expressed proteins (DEPs) between normal and unilateral cryptorchid testes of yaks using high-throughput transcriptome sequencing (RNA-seq) and Tandem Mass Tag (TMT)-based proteomics techniques, with testes from yaks with cryptorchidism as the target. Enrichment analysis revealed that the DEGs were associated with cell growth, sperm motility, immune regulation, and intercellular tight junctions, and were mainly enriched in pathways related to cell differentiation; amino acid, sugar, and lipid metabolism; cell adhesion, and hypoxia tolerance.

This study explores whether the current external quality assessment (EQA) level and acceptable bias for basic semen analysis in China are clinically useful. We collected data of semen EQA from Andrology laboratories in the Hunan Province (China) in 2022 and searched for data in the published literature from January 2000 to December 2023 in China. On the basis of these data, we analyzed the coefficients of variation and acceptable biases of different quality control materials for basic semen analysis through robust statistics.

Purpose: The quantitatively measured sperm DNA damage and disturbed chromatin condensation indexes (%DNA fragmentation index [DFI%] and %high DNA staining index [%HDS]) and their relationships with sperm quality in normospermic men with unexplained infertility were investigated.
