Objective: Thyroid parameters undergo significant dynamic changes during pregnancy. This study aimed to comprehensively analyze the impact of abnormal thyroid parameters in each trimester on the incidence of common adverse obstetric outcomes.
Methods: Blood samples drawn for thyroid parameters in each trimester during the antenatal period were determined after the participants gave birth. Serum thyrotropin, free thyroxine, free triiodothyronine, anti-thyroid peroxidase antibody (TPOAb), and anti-thyroglobulin antibody (TgAb) levels were tested using electrochemiluminescence immunoassays.
Results: Among all the participants, TAI and hypothyroxinemia in the first trimester (T1) were significantly related to an increased risk of gestational hypertension [OR=5.136, 95% CI 1.537-17.158 and OR=7.683, 95% CI: 1.890-31.229, respectively]. Additionally, subclinical hypothyroidism in T1 was independently associated with a higher risk of postpartum hemorrhage [OR = 38.063, 95% CI 2.091-692.834].Besides, subclinical thyrotoxicosis in T1 showed a significant correlation with a raised risk of small for gestational age [OR=14.650, 95% CI 1.221-175.760]. Among euthyroid women during the whole pregnancy, either TPOAb+ or TgAb+ in the third trimester was an independent risk factor of premature birth [OR=5.092, 95% CI 1.059-24.481] and low birth weight [OR=8.165, 95% CI 1.717-38.824], respectively.
Conclusion: Our findings indicate the importance of screening thyroid parameters in early pregnancy and the need to dynamically monitor these parameters throughout the entire pregnancy.

To develop a cell-based in vitro thyroid-stimulating hormone (TSH) biological activity assay that can simulate in vivo pharmacodynamic mechanisms, we constructed two HEK293-TSHR cell lines based on two main cell signaling pathways (Gαs-cAMP-PKA and Gαq/11-PLC-Ca) that TSH depends on for its in vivo physiological function. These cell lines stably expressed the luciferase reporter driven by the cAMP response element (CRE) and nuclear factor of activated T cells (NFAT) response element, and two reporter-gene assays (RGAs) were correspondingly established and validated. The two transgenic genes could measure signals produced from the simulation of the in vivo effects of TSH from the Gαs-cAMP and Gαq/11-PLC pathways after TSH activation.

In the literature, there is no study investigating the relationship between thyroid hormones in pregnancies at 41 weeks and above and the birth timing, labor duration, frequency of fetal distress, premature rupture of membranes (PROM), and maternal hemogram values. A total of 68 nulliparous pregnant women who were admitted to Basaksehir Cam and Sakura City Hospital with indications of delivery between August 2023 and January 2024, between the ages of 20 and 38 and with no comorbidities, were included in the study. Pregnant women with ≥41 weeks of gestation were classified as the late-term pregnancy group ( = 37), and those between 37 and 38 weeks were classified as the control group ( = 31).

Diabetic nephropathy is a severe chronic complication characterized by cytotoxicity, inflammation, and fibrosis, ultimately leading to renal failure. This study systematically investigated the effects of the PARP1 inhibitor PJ-34 on high glucose-induced cytotoxicity, inflammation, and fibrosis in HK-2 cells, as well as its improvement on neuropathic pain response and transforming growth factor β (TGFβ) expression in a type 1 diabetes mellitus diabetic nephropathy mouse model. Through cellular and animal experiments, we observed that PJ-34 significantly enhanced the proliferative capacity of cells damaged by high glucose, reduced apoptosis, and decreased the release of proinflammatory factors TGFα, interleukin-6, and interleukin-1β.
