Three new compounds including two thiodiketopiperazine derivatives shovelmycins A-B (1-2), and one ansamycin derivative divergolide X (3) were isolated and identified from the culture extract of Streptomyces olivaceus HDN22-001, a marine actinomycete obtained from the deep-sea cold seep sediment sample collected from the South China Sea. Their structures and absolute configurations were determined by spectroscopic analyses and ECD calculations. Compound 1 exhibited the strongest DPPH radical scavenging activity with an IC value of 10.83 μM, which was better than that of the positive control vitamin C. And compound 2 was modestly cytotoxic against NCl-H446 cell with the IC value of 26.6 μM.

Moringadepsin () and chaetone B () were isolated by us in the course of a conventional chemical screening of CBS 303.81, a fungus belonging to the relatively underexplored family Schizotheciaceae of the phylum Ascomycota. Using mass spectrometry (MS)-guided isolation methods, a new thiodiketopiperazine derivative (1) and exserohilone (2) were isolated from an EtOAc-extract of Setosphaeria rostrata culture medium. The chemical structure of the new compound was elucidated by MS and NMR spectroscopy, and the absolute configurations were established by the quantum mechanical calculations of electronic circular dichroism. All isolated compounds were examined for their effects on reactive oxygen species (ROS) production, matrix metalloproteinase 1 (MMP-1) secretion, and procollagen type I α1 secretion in tumor necrosis factor (TNF)-α-induced human dermal fibroblasts.

Phosphoglycerate kinase 1 (PGK1) acts as both a glycolytic enzyme and a protein kinase playing critical roles in cancer progression, thereby being regarded as an attractive therapeutic target for cancer treatment. However, no effective inhibitor of PGK1 has been reported. Here, we demonstrate that GQQ-792, a thiodiketopiperazine derivative from marine nature products, is a non-ATP-competitive inhibitor of PGK1 with the disulfide group within the structure of GQQ-792 as a key pharmacophore.
