Chikungunya is a mosquito-borne viral disease that causes fever and severe joint pain for which there is no direct acting drug treatments. Vinyl sulfone SGC-NSP2PRO-1 (3) was identified as a potent inhibitor of the nsP2 cysteine protease (nsP2pro) that reduced viral titer against infectious isolates of Chikungunya and other alphaviruses. The covalent warhead in 3 captured the active site C478 and inactivated nsP2pro with a k/K ratio of 5950 M s. The vinyl sulfone 3 was inactive across a panel of 23 other cysteine proteases and demonstrated remarkable proteome-wide selectivity by two chemoproteomic methods. A negative control analog SGC-NSP2PRO-1N (4) retained the isoxazole core and covalent warhead but demonstrated > 100-fold decrease in enzyme inhibition. Both 3 and 4 were stable across a wide range of pH in solution and upon prolonged storage as solids. Vinyl sulfone 3 and its negative control 4 will find utility as high-quality chemical probes to study the role of the nsP2pro in cellular studies of alphaviral replication and virulence.

Mas-related G protein-coupled receptor X2 (MrgX2) plays a key role in pseudoallergy reactions; thus, it is of great significance to screen compounds with antipseudoallergy activity via MrgX2. Cell membrane chromatography (CMC) demonstrates great potential in drug screening, but it requires further optimization to improve its specificity and stability. In this study, a new CMC system incorporating His-tag-oriented immobilized proteins was constructed to screen MrgX2 antagonists.

In this report we try to answer the question if smart proteolytic samplers can be used for LC-MS based quantitative determination of proteins from paper. Production of smart proteolytic samplers is a two-stage process: In the first stage cellulose is functionalized while in the second stage trypsin is covalently bound. Two functionalization strategies, divinyl sulfone - and periodate functionalization, were compared under various conditions.

Periodontitis is a chronic inflammatory condition of the periodontium, which often leads to tooth loss. Recently, statins have emerged as potent anti-inflammatory agents with pleiotropic effects that can potentially outperform conventional periodontal treatments. However, the clinical application of statins is limited by the lack of suitable drug carriers that fit the periodontal region and provide a controlled local drug release.

Supercritical water gasification (SCWG) is famous for the clean utilization of organic wastes without SO emission. Investigating the decomposition mechanism of sulfone compounds, the dominant organic sulfur compounds of organic wastes, in supercritical water (SCW) is conducive to the development of SCWG technology. Herein, the comparative decomposition mechanism of phenyl vinyl sulfone (PVS), diphenyl sulfone (DS), and benzo[b]thiophene 1,1-dioxide (BD) are explored via experiments and density functional theoretical (DFT) calculations.
