Structured RNAs play a crucial role in regulating gene expression, which includes both protein synthesis and RNA processing. Dysregulation of these processes is associated with various conditions, including viral and bacterial infections, as well as cancer. The unique tertiary structures of structured RNAs provide an opportunity for small molecules to directly modulate such processes, making them promising targets for drug discovery. Although small-molecule inhibitors targeting RNA have shown early success, strategies like structure-based virtual screening remain underutilized for RNA-targeted drug discovery. In this study, we developed a virtual screening scheme targeting the structural ensemble of EV-A71 IRES SL II, a noncoding viral RNA element essential for viral replication. We subsequently optimized the experimentally validated hit compound IRE-03 from virtual screening through an "analog-by-catalog" search. This led to the identification of a more potent IRES inhibitor, IRE-03-3, validated through biochemical and functional assays with an EC value of 11.96 μM against viral proliferation. Our findings demonstrate that structure-based virtual screening can be effectively applied to RNA targets, providing exciting new opportunities for future antiviral drug discovery.

Background: The receptor-binding domain (RBD) of the spike protein of SARS-CoV-2 interacts with the angiotensin-converting enzyme 2 (ACE2) receptor in humans. To date, numerous SARS-CoV-2 variants, particularly those involving mutations in the RBD, have been identified. These variants exhibit differences in transmission, pathogenicity, diagnostics, and vaccine efficacy.

, a versatile and antibiotic-resistant gram-negative pathogen, poses a critical threat to both immunocompromised and immunocompetent populations, underscoring the urgent need for new therapeutic targets. This study applies an extensive subtractive proteomics approach to identify viable drug targets within the core proteome of , analyzing a total of 5563 proteins. Through a rigorous, multi-stage process, we excluded human homologs, identified essential proteins, mapped functional pathways, determined subcellular localization, and assessed virulence and resistance factors.

Background: Up to 75% of young adult cancer survivors (YACS) experience chronic insomnia, negatively affecting physical and emotional health and overall quality of life. Cognitive behavioral therapy for insomnia (CBT-I) is a gold-standard intervention to address insomnia. To improve CBT-I access and treatment adherence, screen-based digital CBT-I platforms have been developed.

The integration of gaze/eye tracking into virtual and augmented reality devices has unlocked new possibilities, offering a novel human-computer interaction (HCI) modality for on-device extended reality (XR). Emerging applications in XR, such as low-effort user authentication, mental health diagnosis, and foveated rendering, demand real-time eye tracking at high frequencies, a capability that current solutions struggle to deliver. To address this challenge, we present EX-Gaze, an event-based real-time eye tracking system designed for on-device extended reality.

The paper describes SeamlessVR, a method for switching effectively from immersive visualization, in a virtual reality (VR) headset, to non-immersive visualization, on screen. SeamlessVR implements a continuous morph of the 3D visualization to a 2D visualization that matches what the user will see on screen after removing the headset. This visualization continuity reduces the cognitive effort of connecting the immersive to the non-immersive visualization, helping the user continue on screen a visualization task started in the headset.
