Palladium-Catalyzed Insertion of Allenes into Dithioacetals To Access 1,3-Dithioethers.

Great efforts have been devoted to synthesizing dithioethers due to their unique pharmaceutical properties. While significant success has been achieved in the synthesis of 1,1-dithioethers and 1,2-dithioethers, a concise and efficient strategy for the direct construction of 1,3-dithioethers has not yet been established. Herein, we developed a palladium-catalyzed thiomethylthiolation of allenes with dithioacetals to access a wide range of 1,3-dithioethers through the simultaneous construction of one carbon-carbon bond and one carbon-sulfur bond. A dithioacetal-coordinated cationic palladium complex was isolated and characterized, which was considered the crucial intermediate for this palladium-catalyzed transformation.

The mechanism and origin of selectivities in BF3·Et2O-catalyzed intermolecular [3 + 2] cycloadditions of propargylic alcohol and α-oxo ketene dithioacetals have been studied using density functional theory. Several possible reaction pathways were evaluated on the basis of two possible binding modes between the carbonyl or hydroxyl oxygen of substrates and catalyst. The preferred mechanism initiates dehydroxylation of propargylic alcohol by Lewis acid BF3 and generates active allenic carbocation species to provide the favorable electrophile.

The cycloisomerization reactions of allenes bearing cyclic acetal, thioacetal and dithioacetal subunits, when triggered either by the catalytic action of AgSbF6 or by one equiv. of CF3COOH, gave rise to four different classes of indeno-fused 1,4-dioxa, oxathia and dithia heterocycles, in most cases as a single diastereomer. Acyclic acetals and dithioacetals are also suitable starting materials in similar transformations yielding 1,2-disubstituted indenes and 1,3-disubstituted 2-alkylideneindanes.

A convenient base-mediated strategy to synthesize 3-aryol-4-methyl (or benzyl)-2-methylthio furans 2 (trisubstituted furans) has been developed through the domino coupling/annulations between available α-oxo ketene dithioacetals 1 and propargyl alcohols. In this strategy, these types of bases play an important role in driving the domino coupling reaction of propargyl alcohols and further intramolecular annulations to realize the target compounds. The possible mechanism for the formation of the various products is believed to involve the generation of allenes 7, followed by intramolecular annulations.

Synthesis and Synthetic Application of Phosphonoketene Dithioacetals. New Synthesis of Dithioallenes and (alpha-Dithiocarboxyvinyl)phosphonates.

Phosphonoketene dithioacetals 3a-e were obtained in good yields by the reaction of ethyl phosphonoacetates 1a,b with 2-4 equiv of thiols 2a-c in the presence of an alkylaluminum dichloride or dialkylaluminum chlorides. Reaction of 2,2-dithio-1-phosphonovinyl anions with aldehydes afforded allylic alcohols 4-7, 11-18 in good to moderate yields. Treatment of the alcohols 4-6 with t-BuOK in THF led to symmetrical [2 + 2] cycloadducts 20-22 of 1,1-(ethylenedithio)allenes in moderate yields, while a similar reaction of the alcohols 11-13 produced a mixture of symmetrical and unsymmetrical [2 + 2] cycloadducts of 1,1-(trimethylenedithio)allenes,23a-25a and 23b-25b, in 55-94% yields.
