Biomotor-driven DNA packaging is a key step in the life cycle of many viruses. We previously developed single-molecule methods using optical tweezers to measure packaging dynamics of the bacteriophage lambda motor. The lambda system is more complex than others examined via single-molecule assays with respect to the packaging substrate and ancillary proteins required. Because of this, previous studies which efficiently detected packaging events used crude E. coli cell extracts containing host factors and the terminase packaging enzyme. However, use of extracts is suboptimal for biochemical manipulation and obfuscates interrogation of additional factors that affect the process. Here we describe an optical tweezers assay using purified lambda terminase holoenzyme. Packaging events are as efficient as with crude extracts, but only if purified E. coli integration host factor (IHF) is included in the motor assembly reactions. We find that the ATP-driven DNA translocation dynamics, motor force generation, and motor-DNA interactions without nucleotide are virtually identical to those measured with extracts. Thus, single-molecule packaging activity can be fully recapitulated in a minimal system containing only purified lambda procapsids, purified terminase, IHF, and ATP. This sets the stage for single-molecule studies to investigate additional phage proteins known to play essential roles in the packaging reaction.

This study investigates developing and characterizing electrospun nanofibers composed of polyvinyl alcohol (PVA) and oxidized xanthan gum (OXG), with nisin as a bioactive agent, for innovative food packaging applications. Fourier-transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM) confirmed successful crosslinking between PVA and OXG, along with uniform nisin dispersion within the fibers. The inclusion of OXG increased moisture content (MC) and water solubility (WS) while reducing porosity and water vapor permeability (WVP), demonstrating its role as a crosslinker.

Traditional wood-plastic composites (WPC) face numerous challenges in their applications, such as the lack of flexibility, cushioning performance, water barrier properties, biodegradability and recyclability. To address these issues, in this work, a recyclable lignocellulosic biocomposites with excellent comprehensive performance was developed using cork flour and poly(1,4-dioxan-2-one) (PPDO) as the green raw materials. The prepared cork/PPDO WPC contained up to 70 wt% cork flour, conferring them with enhanced flexibility (elongation at break >10 %) and cushioning performance due to the strong interactions between PPDO and cork flour, as well as the cork unique flexibility from cork cavity structure and suberin.

To address the challenges associated with the storage and application of traditional carbon dot (CDs) solutions, this study introduces a cyan fluorescent carbon dot-based hydrogel (CDs-SCH). The hydrogel was synthesized by integrating cyan fluorescent CDs, derived from penicillamine and m-phenylenediamine, with carboxymethylcellulose (CMC) and sodium alginate (SA), which was then mixed with acrylamide (AM). The resulting CDs-SCH hydrogel was extensively characterized, focusing on its morphology, chemical structure, and fluorescence behavior.
