T-21 is a lactic acid bacterium isolated from wild cranberries in Japan that demonstrates significant immunomodulatory properties and has been incorporated into commercial health products. However, probiogenomic analyses specific to T-21 have remained largely unexplored. This study performed a thorough genomic characterisation of T-21 and evaluated its safety in initial clinical trials. Genomic analysis revealed substantial genetic diversity and metabolic capabilities, including enhanced fermentative potential demonstrated by its ability to metabolise a wide range of plant-derived carbohydrates, and genetic determinants associated with exopolysaccharide biosynthesis and nisin production, distinguishing T-21 from domesticated dairy strains. These attributes, reflective of its wild plant origin, may contribute to its metabolic versatility and unique probiotic functionalities. A preliminary clinical trial assessing the safety of T-21-fermented milk in healthy Japanese adults indicated no significant adverse outcomes, corroborating its safety for human consumption. Together, these findings support the feasibility of utilising non-dairy, wild plant-origin strains in dairy fermentation processes as probiotics. This study expands our understanding of the genomic basis for T-21's probiotic potential and lays the groundwork for further investigations into its functional mechanisms and potential applications in promoting human health.

Aim: To characterize the deep dentinal caries microbiota in teeth diagnosed with normal pulp with deep caries (NP), reversible pulpitis (RP), symptomatic irreversible pulpitis (SIP), and asymptomatic irreversible pulpitis (AIP), and to identify potential key pathogens associated with pulpitis progression, exploring their roles in disease advancement.
Methodology: In this cross-sectional study, we collected 108 dentinal caries samples, categorized into NP (n = 27), RP (n = 27), SIP (n = 27), and AIP (n = 27), according to the American Association of Endodontists' diagnostic criteria. 2 NP samples and 2 RP samples were excluded due to contamination.

Unlabelled: Insertion sequences (ISs) are key components of most bacterial genomes and play a crucial role in bacterial mutagenesis. In this study, we observed the insertion of an IS element, ISLrh, from the M1 genome into plasmid pGK12, resulting in the generation of a new plasmid, pTRK829. This insertion enabled pTRK829 to replicate in hosts previously incompatible with pGK12, including M1, GG (LGG), ATCC 393, and ATCC 25598.

Growth differentiation factor 15 (GDF15) is upregulated in most cases of epithelial ovarian cancer (EOC); however, its functions in EOC are not fully understood. In this study, we knocked down GDF15 in EOC cells before performing high-throughput sequencing to identify genes regulated by GDF15. GDF15 was overexpressed to determine its effect on the viability, migration, and response of EOC cells to gemcitabine, carboplatin, and paclitaxel.

The construction of functional micro- or nanostructured surfaces is extensively studied since they are able to provide multifunctional properties and for large variety of potential applications in fields such as tissue engineering, wearable electronics or microfluidics.
