Introduction: Neuropsychiatric symptoms in dementia (NPS) collectively refer to behavioral and psychological symptoms affecting individuals with mild cognitive impairment (MCI) or Alzheimer's disease or related dementia (ADRD). NPS are among the most troubling aspects of living with dementia and their treatments have limited efficacy. We aim to investigate genetic variants contributing to NPS to identify new therapeutic targets.
Methods: We performed a genome-wide association study (GWAS) for nine NPS domains measured by the NPI-Q in 12,800 participants of European ancestry with MCI or ADRD recruited by Alzheimer's disease research centers across the U.S.
Results: We found genome-wide significant signals for agitation, anxiety, apathy, delusions, and hallucinations in the locus that were driven by the ε4 allele. We replicated these findings in two independent datasets. Mediation analyses revealed that MCI/ADRD severity only partially mediated the GWAS signals, except for apathy.
Discussion: These findings suggest the ε4 allele influences NPS independently of and beyond its effect on ADRD.

Chromosome 22q11.2 deletion increases the risk of neuropsychiatric disorders like autism and schizophrenia. Disruption of large-scale functional connectivity in 22q11 deletion syndrome (22q11DS) has been widely reported, but the biological factors driving these changes remain unclear.

Importance: Peripheral (blood-based) biomarkers for psychiatric illness could benefit diagnosis and treatment, but research to date has typically been low throughput, and traditional case-control studies are subject to potential confounds of treatment and other exposures. Large-scale 2-sample mendelian randomization (MR) can examine the potentially causal impact of circulating proteins on neuropsychiatric phenotypes without these confounds.
Objective: To identify circulating proteins associated with risk for schizophrenia (SCZ), bipolar disorder (BD), and major depressive disorder (MDD) as well as cognitive task performance (CTP).

Neurochemical imbalance is a contributing factor to neurological symptoms in multiple sclerosis (MS). The matured myelin sheath is crucial for substance transportation within the extracellular space (ECS) and for maintaining local homeostasis. Therefore, we hypothesize that disturbed ECS transportation following demyelinating lesions might lead to neurochemical imbalance in MS.

Objective: Safety net enclosures are used in addition to (non-) pharmacological interventions in patients suffering from neuropsychiatric symptoms in dementia. However, no data on effectiveness are available.
Methods: In a prospective observational cohort study of 81 patients diagnosed with dementia, and admitted to a geriatric ward of a psychiatric hospital, available behavioral assessment scores were used to compare 45 patients who used safety net enclosures with 36 patients who never used safety net enclosures.

Background: Depression is a widely recognized neuropsychiatric condition that often occurs as a comorbidity with various medical illnesses, including neurodegenerative disorders like Parkinson's disease (PD). This study aimed to identify the age of onset and underlying disease characteristics associated with patients exhibiting mild to moderate depression comorbid with PD.
Methods: This retrospective case-control study included 114 elderly patients (age ≥65 years) diagnosed with Parkinson's disease.
