NAD(P)H:quinone oxidoreductase-1 (NQO1), a protein highly expressed in tumor cells, serves as an excellent trigger for releasing drugs specifically within tumor cells. In this study, we designed an activatable circular antisense oligonucleotide (cASO) by incorporating a head-to-tail cyclization mediated by an NQO1-responsive trimethyl-locked quinone propionate (Q3PA), coupled with a self-immolative linker. The resulting circular structure prevented the cASO from binding to the target mRNA, thereby avoiding gene silencing. However, upon encountering NQO1, the circular form was converted to a linear form, leading to the silencing of the targeted gene. experiments demonstrated significant tumor-cell-specific activity of the cASO, while studies using an A549-Luc orthotopic lung tumor model revealed a substantial antitumor effect, primarily attributed to the suppression of survivin expression. This NQO1-activatable cASO represents a novel strategy for achieving tumor-cell-specific gene silencing and holds promise for the development of ASO prodrugs with enhanced therapeutic potentials.

The vaccinia virus Tiantan (VTT) is widely utilized as a smallpox vaccine in China and holds significant importance in the prevention of diseases stemming from poxvirus infections. Nevertheless, few studies have investigated the influence of VTT infection on host gene expression. In this study, we constructed time series transcriptomic profiles of HeLa cells infected with both VTT and western reserve (WR) strains.

Although the significance of some plant WRKYs in response to cold stress have been identified, the molecular mechanisms of most WRKYs remain unclear in grapevine. In this study, we demonstrate that cold-induced expression of in executes a beneficial role in enhancing resistance by the regulating starch decomposition. VaWRKY65 was identified as an upstream transcriptional activator of through yeast one-hybrid library screening and validated to directly interact with the W-box region inside the promoter.

Brassinosteroids (BRs) are extensively distributed in plants and play crucial roles throughout all stages of plant growth. Nevertheless, the molecular mechanism through which BRs influence postharvest senescence in pakchoi remains elusive. Previous studies have demonstrated that the application of 1.
