Bone is subjected to a plethora of mechanical stresses, which have been found to directly influence the equilibrium between bone resorption and formation. Taking this into account, we present herein a novel biomimicking 3-dimensional model that applies cyclic uniaxial compression onto cells co-cultured on 3-dimensionally printed scaffolds consisting of poly L-lactic acid/poly(ε-caprolactone)/poly(3-hydroxybutyrate--3-hydroxyvalerate)/Sr-nanohydroxyapatite. The aim is to investigate how compression can modulate the balance between osteogenesis and osteoclastogenesis in co-culture, as well as the polarization of macrophages.

Impaired branched-chain amino acid (BCAA) catabolism during adipocyte differentiation decreases glycolytic flux.

Dysregulated branched-chain amino acid (BCAA) metabolism has emerged as a key metabolic feature associated with the obese insulin-resistant state, and adipose BCAA catabolism is decreased in this context. BCAA catabolism is upregulated early in adipogenesis, but the impact of suppressing this pathway on the broader metabolic functions of the resultant adipocyte remains unclear. Here, we use CRISPR/Cas9 to decrease BCKDHA in 3T3-L1 and human pre-adipocytes, and ACAD8 in 3T3-L1 pre-adipocytes to induce a deficiency in BCAA catabolism through differentiation.

Exosomal SLC16A1-AS1-induced M2 macrophages polarization facilitates hepatocellular carcinoma progression.

Int J Biol Sci

September 2024

Department of Emergency Surgery, Union Hospital, Tongji Medical College, Huazhong University of Science and Technology, Wuhan 430022, China.

Article Synopsis
  • Macrophages play a crucial role in the tumor microenvironment (TME) and communicate with tumor cells, significantly influencing tumor growth and development.
  • The study focuses on exosomal lncRNAs, particularly SLC16A1-AS1 from hepatocellular carcinoma (HCC) exosomes, which drives the malignant progression by promoting M2 polarization of macrophages through a specific molecular mechanism.
  • The interaction between SLC16A1-AS1 and M2 macrophages enhances HCC cell behaviors like proliferation and invasion, suggesting that targeting SLC16A1-AS1 could be a potential therapeutic strategy against HCC.
