An enzyme with strong single-stranded DNA (ssDNA) ligation activity would be advantageous for many molecular biology applications. However, currently available enzymes exhibit only limited activity. Here, we identified an enzyme with strong ssDNA ligation activity upon searching the databases for proteins homologous to TS2126 RNA ligase, the known enzyme with the highest yet limited ssDNA ligation activity. A ligase from Thermus phage phiLo (GenBank: AYJ73970) or Tph ssDNA ligase (SDL) depicted higher ssDNA ligation activity than TS2126 RNA ligase. This study suggests that SDL could be useful for various applications including sequencing for library preparation.

Recent studies have shown that anti-ERp5 antibodies inhibit platelet activation and thrombus formation; Moreover, ERp5-deficient platelets exhibit enhanced platelet reactivity via regulation of endoplasmic reticulum (ER) stress. In this study, we used a new ERp5-knockout mouse model as well as recombinant ERp5 (rERp5) protein, to examine the role of ERp5 in platelet function and thrombosis. Although platelet-specific ERp5-deficient mice had decreased platelet count, the mice had shortened tail-bleeding times and enhanced platelet accumulation in FeCl-induced mesenteric artery injury, compared with wild-type mice.

Engineered heme proteins possess excellent biocatalytic carbene N-H insertion abilities for sustainable synthesis, and most of them have His as the Fe axial ligand. The ligation strategy has been widely used in the chemical synthesis of atomically precise clusters. A series of thymine (T)-ligated Al-T ( = Be, Al, C; = 1-5) complexes have been studied to reveal the effect of DNA nucleobase ligands on the electronic structures of different superatoms in the present work. In addition to its protective role, the successive attachment of thymine ligands significantly lowers the adiabatic ionization energies (AIEs) of the studied Al superatoms with filled and unfilled electronic shells.
