Globally, many complex issues, like the ageing population and health inequalities, require attention. People are experimenting to combat these issues in their local contexts through bigger or smaller networks; however, much of the knowledge about these initiatives remains localised and elitist and omits the voices and perspectives of citizens. This article identifies the characteristics of a more horizontal, emergent and plural epistemology to mobilize knowledge. We used local networks building age-friendly communities in the Netherlands as a case study. With members of 110 local networks, we worked with a new methodology called to create fruitful interactions and learn with stakeholders, including older citizens, in a learning community. Four working principles helped us, namely (1) a rhizomatic design based on multiplicity, heterogeneity and non-linearity of knowledge; (2) fertile soil nurtured by the talents and wisdom of participants through participatory arts-based methods; (3) so-called 'wicked skills' of a forester 2.0 to facilitate learning; and (4) an ecosystem metaphor as a boundary object to understand local networks. We invite colleagues to experiment with dynamic knowledge synthesis to connect on different levels, with particular attention to the inclusion of citizens in creating sustainable local communities.

Cardiomyocytes can be implanted to remuscularize the failing heart. Challenges include sufficient cardiomyocyte retention for a sustainable therapeutic impact without intolerable side effects, such as arrhythmia and tumour growth. We investigated the hypothesis that epicardial engineered heart muscle (EHM) allografts from induced pluripotent stem cell-derived cardiomyocytes and stromal cells structurally and functionally remuscularize the chronically failing heart without limiting side effects in rhesus macaques.

Collective behavior in biological systems emerges from local interactions among individuals, enabling groups to adapt to dynamic environments. Traditional modeling approaches, such as bottom-up and top-down models, have limitations in accurately representing these complex interactions. Attention mechanisms such as the Convolutional Block Attention Module (CBAM) can help emphasize and refine the most relevant feature maps such as color, texture, spots, and wrinkle variations for the avocado ripeness classification. However, the CBAM lacks global context awareness, which may prevent it from capturing long-range dependencies or global patterns such as relationships between distant regions in the image. Further, more complex neural networks can improve model performance but at the cost of increasing the number of layers and train parameters, which may not be suitable for resource constrained devices.

Isocitrate dehydrogenase 1/2 mutant (IDHmt) astrocytoma is considered a T cell-deprived tumor, yet little is known regarding the phenotypes underlying T cell exclusion. Using bulk, single nucleus and spatial RNA and protein profiling, we demonstrate that a distinct spatial organization underlies T cell confinement to the perivascular space (T cell cuff) in IDHmt astrocytoma. T cell cuffs are uniquely characterized by a high abundance of gemistocytic tumor cells (GTC) in the surrounding stroma.

Blood clots (BCs) play a crucial biomechanical role in promoting osteogenesis and regulating mesenchymal stem cell (MSC) function and fate. This study shows that BC formation enhances MSC osteogenesis by activating Itgb1/Fak-mediated focal adhesion and subsequent Runx2-mediated bone regeneration. Notably, BC viscoelasticity regulates this effect by modulating Runx2 nuclear translocation.
