The paper is concerned with experimental and clinical problems. Posttranslation modification of ferritin with glucose has been shown to cause change of its immunochemical properties. Changes in iron metabolism and in the level of glycosylated hemoglobin A1c were noted in patients with different forms of diabetes mellitus.

Skeletal muscles account for ~30-40% of the total weight of human body and are responsible for its most important functions, including movement, respiration, thermogenesis, and glucose and protein metabolism. Skeletal muscle damage negatively impacts the whole-body functioning, leading to deterioration of the quality of life and, in severe cases, death. Therefore, timely diagnosis and therapy for skeletal muscle dysfunction are important goals of modern medicine.

IgA class switching enhances neutralizing potency against SARS-CoV-2 by increased antibody hinge flexibility.

IgA antibodies are critical components of the mucosal immune barrier, providing essential first-line defense against viral infections. In this study, we investigated the impact of antibody class switching on neutralization efficacy by engineering recombinant antibodies of different isotypes (IgA1, IgG1) with identical variable regions from SARS-CoV-2 convalescent patients. A potent, broad-spectrum neutralizing monoclonal antibody CAV-C65 exhibited a ten-fold increase in neutralization potency upon switching from IgG1 to IgA1 monomer.

A general temperature-guided language model to design proteins of enhanced stability and activity.

November 2024

School of Physics and Astronomy, & Shanghai National Center for Applied Mathematics (SJTU Center), & Institute of Natural Sciences, Shanghai Jiao Tong University, Shanghai 200240, China.

Designing protein mutants with both high stability and activity is a critical yet challenging task in protein engineering. Here, we introduce PRIME, a deep learning model, which can suggest protein mutants with improved stability and activity without any prior experimental mutagenesis data for the specified protein. Leveraging temperature-aware language modeling, PRIME demonstrated superior predictive ability compared to current state-of-the-art models on the public mutagenesis dataset across 283 protein assays.

Article Synopsis
  • Laser-induced breakdown spectroscopy (LIBS) enhances immunochemical assays using indirect label detection with nanoparticles, specifically yttrium-based photon-upconversion nanoparticles (UCNPs), achieving sensitivity comparable to established methods like fluorescence immunoassays.
  • The study improves the sensitivity of UCNP-based Tag-LIBS by incorporating magnetic microbeads (MBs), which allow for analyte preconcentration and significantly lowers the limit of detection (LOD) to 4.0 pg/mL, a notable advancement over traditional microtiter plate methods.
  • The technique demonstrates robust performance in clinical samples and shows strong correlation with standard assays, indicating that MB-based Tag-LIBS offers fast readout, high sensitivity, and potential for multiplexing without relying
