Bioeffects of Nanoplastics: DNA Damage and Mechanism.

Nanoplastics, as emerging contaminants, have been causing great panic, potentially affecting human health in recent years. Some studies have indicated that nanoplastics may induce severe toxicity. However, the mechanisms underlying this potential toxicity are insufficiently understood. In this study, we have found PS-NH nanoplastics had the obvious DNA cleavage activities, while PS-COOH nanoplastics were not observed to have the DNA cleavage abilities. Both microsized PS-NH and PS-COOH microplastics lacked DNA cleavage activities, indicating the importance of size and surface ligand in nanoplastics' DNA cleavage. The DNA cleavage system by nanoplastics remains stable under varying pH and temperature. From the mechanism exploration, the interaction energy is much higher between PS-NH nanoplastics than PS-COOH nanoplastics, further illuminating that PS-NH nanoplastics have stronger binding interaction with DNA to induce DNA cleavage activities. This study offers insights into the potential environmental risks and toxicity of nanoplastics in the aquatic ecosystems.

As a promising therapeutic approach, the RNA editing process can correct pathogenic mutations and is reversible and tunable, without permanently altering the genome. RNA editing mediated by human ADAR proteins offers unique advantages, including high specificity and low immunogenicity. Compared to CRISPR-based gene editing techniques, RNA editing events are temporary, which can reduce the risk of long-term unintended side effects, making off-target edits less concerning than DNA-targeting methods.

Colorectal cancer (CRC) is a leading cause of cancer-related deaths globally, necessitating the development of sensitive and minimally invasive diagnostic approaches. In this study, we present a novel diagnostic strategy by integrating dumbbell probe-mediated CRISPR/Cas13a with nicking-induced DNA cascade reaction (DP-bridged Cas13a/NDCR) for highly sensitive microRNA (miRNA) detection. Target miRNA triggers Cas13a-mediated cleavage of the dumbbell probe, releasing an intermediate strand that hybridizes with a methylene blue-labeled hairpin probe on the electrode surface.
