Sprayable hydrogels have emerged as a transformative innovation in biomedical technology, offering a versatile, efficient, and minimally invasive platform for various clinical applications. They form gels upon tissue contact, enabling seamless application on even complex surfaces. This property is especially useful in wound care, drug delivery, and tissue engineering, where localized and sustained release of therapeutics is essential. Formulations can be customized to include various bioactive compounds, such as growth factors, antibiotics, and anti-inflammatory agents, thereby enhancing targeted treatment outcomes. This review delves into the fundamental principles governing sprayable hydrogels, emphasizing critical mechanisms such as cross-linking, shear-thinning properties, and thermoresponsive behavior. Furthermore, it highlights recent advancements since 2020, including the strategic incorporation of bioactive agents to augment therapeutic efficacy. By examining these core mechanisms and design strategies, this review provides a comprehensive perspective on the engineering of sprayable hydrogels for modern medical applications.

While bipolar disorder patients can benefit from lithium therapy, high levels of lithium in the serum can induce undesirable systemic side effects. Intranasal (IN) lithium delivery offers a potential solution to this challenge given its potential to facilitate improved lithium transport to brain when delivered to the olfactory mucosa. Herein, a sprayable, in situ forming nanoparticle network hydrogel (NNH) based on Schiff base interactions between chelator-functionalized oxidized starch nanoparticles (SNPs) and carboxymethyl chitosan (CMCh) is reported that can be deployed within the nasal cavity to release ultra-small penetrative SNPs over time.

To address the issues of infectious virus, bacterial secondary infections, skin pigmentation, and scarring caused by monkeypox virus (MPXV), a sprayable hydrogel with versatile functions was developed with comprehensive properties. Based on current research, the bioactive deep eutectic solvent (DES) of rosmarinic acid-proanthocyanidin-glycol (RPG) was designed and synthesized as active agent, and molecular docking was applied to discover its binding to MPXV proteins through H-bonds and van der Waals interactions, and the docking results show the binding energies between RA, PC, Gly and MPXV proteins are -58.7188, -50.

Necrosis at the distal end of random skin flaps remains a significant challenge, limiting the clinical application of these flaps in plastic and reconstructive surgery. Inhibiting ischemia/reperfusion (I/R) injury and promoting the formation of neovascular networks are critical preventive strategies. Platelet-derived small extracellular vesicles (PL-sEV) are nanocarriers of growth factors that provide an alternative to clinically used platelet-rich plasma and platelet lysates, offering higher growth factor concentrations and lower immunogenicity.
