DegSU quorum sensing (QS) system enables autoinducible expression of recombinant proteins in . However, insufficient promoter strength and a complex regulatory circuit limit its practical application. Here, the QS-responsive promoter P was modified by core region mutation, upstream truncation, and addition of activating binding sites, yielding P with a 118.3% increase in strength. A mathematical model was developed to accurately quantify the regulatory process from a comprehensive perspective. Guided by this model, the DegSU QS system was further optimized in a robust by knocking out competitive target genes and , operons and , introducing variants and 36, and increasing regulatory strength by 84.0%. A 52.5% increase in acetoin titer and a 65.9% increase in extracellular carboxypeptidase activity validated the industrial value of this study. Overall, this study addresses the limitations of the DegSU QS system in practical application and demonstrates its potential for high-level recombinant protein production.

Bacterial gene expression systems play a crucial role in producing valuable biological macromolecules, such as recombinant proteins and polysaccharides. However, traditional inducible gene systems have limitations that need costly chemical inducers that can harm the host. To address these challenges, a novel peptide-activated auto-inducible gene expression system was developed in Bacillus subtilis, leveraging Accessory gene regulatory system (Agr), a two-component signal system, from Staphylococcus aureus to trigger gene expression in response to an auto-inducible peptide (AIP).

Delaying production with prokaryotic inducible expression systems.

Centre for Synthetic Biology, Ghent University, Coupure Links 653, 9000, Ghent, Belgium.

Background: Engineering bacteria with the purpose of optimizing the production of interesting molecules often leads to a decrease in growth due to metabolic burden or toxicity. By delaying the production in time, these negative effects on the growth can be avoided in a process called a two-stage fermentation.

Main Text: During this two-stage fermentation process, the production stage is only activated once sufficient cell mass is obtained.

Indigo is widely used in textile industries for denim garments dyeing and is mainly produced by chemical synthesis which, however, raises environmental sustainability issues. Bio-indigo may be produced by fermentation of metabolically engineering bacteria, but current methods are economically incompetent due to low titer and the need for an inducer. To address these problems, we first characterized several synthetic promoters in E.

Chemokines are small, secreted proteins with chemoattractive properties, which play an important role in the recruitment and activation of immune cells. CXCL11 is a CXC chemokine specific for the CXCR3 receptors, which has been shown to mediate the generation of Th1-type immune responses and have bactericidal effects similar to defensins. Herein, we cloned the full-length cDNA of Chinese soft-shelled turtle (Pelodiscus sinensis) CXCL11, designated as PsCXCL11, which consist of an open reading frame (ORF) of 282 bp encoding 93 amino acids, with estimated molecular weight of 10.
