Most bacteria lack membrane-enclosed organelles and rely on macromolecular scaffolds at different subcellular locations to recruit proteins for specific functions. Here, we demonstrate that the optogenetic CRY2-CIB1 system from Arabidopsis thaliana can be used to rapidly direct proteins to different subcellular locations with varying efficiencies in live Escherichia coli cells, including the nucleoid, the cell pole, the membrane, and the midcell division plane. Such light-induced re-localization can be used to rapidly inhibit cytokinesis in actively dividing E. coli cells. We further show that CRY2-CIBN binding kinetics can be modulated by green light, adding a new dimension of control to the system. Finally, we test this optogenetic system in three additional bacterial species, Bacillus subtilis, Caulobacter crescentus, and Streptococcus pneumoniae, providing important considerations for this system's applicability in bacterial cell biology.

The spatial transcriptomics is a rapidly evolving biological technology that simultaneously measures the gene expression profiles and the spatial locations of spots. With progressive advances, current spatial transcriptomic techniques can achieve the cellular or even the subcellular resolution, making it possible to explore the fine-grained spatial pattern of cell types within one tissue section. However, most existing cell spatial clustering methods require a correct specification of the cell type number, which is hard to determine in the practical exploratory data analysis.

Background: APOE is the major genetic risk factor for sporadic Alzheimer's disease (AD). Although APOE is well known to promote Aβ pathology, recent data also support an effect of APOE polymorphism on phosphorylated Tau (pTau) pathology.
Method: To elucidate this potential effect, the pTau interactome was analyzed across APOE genotypes in the frontal cortex of 10 advanced AD cases (n = 5 APOE and n = 5 APOE), using a combination of anti-pTau PHF1 (pS396/pS404) immunoprecipitation and mass spectrometry.

The ABORTED MICROSPORES (AMS) gene is crucial for tapetal cell development and pollen formation, but its role in Upland cotton (Gossypium hirsutum) has not been previously documented. This study identified GhAMS11 as a key transcription factor, with its high expression specifically observed during the S4-S6 stages of anther development, a critical period for tapetal activity and pollen formation. Subcellular localization confirmed that GhAMS11 was located in the nucleus.
