Article Synopsis
  • This study investigates the levels of folate receptor autoantibody (FRAA) in children with autism spectrum disorder (ASD) compared to typically-developing (TD) children, finding lower levels of FRAA in the ASD group, particularly among boys.
  • Researchers also assessed serum levels of folate, vitamin B, and ferritin, revealing no significant differences in folate levels between the two groups, but noted specific differences in boys.
  • The study suggests the potential diagnostic value of combining nitrite and binding-FRAA for ASD, highlighting that low FRAA levels in autistic children may be influenced by various factors, warranting further research.

The cerebral folate receptor alpha (FRα) transports 5-methyltetrahydrofolate (5-MTHF) into the brain; low 5-MTHF in the brain causes cerebral folate deficiency (CFD). CFD has been associated with autism spectrum disorders (ASD) and is treated with -leucovorin (folinic acid). One cause of CFD is an autoantibody that interferes with the function of the FRα.

Folic acid has been identified to be integral in rapid tissue growth and cell division during fetal development. Different studies indicate folic acid's importance in improving childhood behavioral outcomes and underline its role as a modifiable risk factor for autism spectrum disorders. The aim of this systematic review is to both elucidate the potential role of folic acid in autism spectrum disorders and to investigate the mechanisms involved.

Background: Supplementation of folic acid by pregnant mothers is thought to lower the risk of autism spectrum disorders (ASDs) in the offspring. Folic acid is taken up by cells via receptors with high affinity for folate and reduced folic acid derivatives. However, this is blocked by the presence of folate receptor autoantibodies (FRAA).
