The interplay between metabolic pathways and the epigenome is essential for proper cell differentiation. In this new study, Francisco Naya and colleagues find that the Dlk1-Dio3 noncoding RNA (ncRNA) locus regulates cell state by coordinating mitochondrial activity and histone modifications in muscle cells. To find out more about the people behind the work, we caught up with first author Amanda Pinheiro and corresponding author Francisco (Frank) Naya, Associate Professor at the Department of Biology, Boston University, USA.

The coordinate regulation of metabolism and epigenetics to establish cell state-specific gene expression patterns during lineage progression is a central aspect of cell differentiation, but the factors that regulate this elaborate interplay are not well-defined. The imprinted Dlk1-Dio3 noncoding RNA (ncRNA) cluster has been associated with metabolism in various progenitor cells, suggesting it functions as a regulator of metabolism and cell state. Here, we directly demonstrate that the Dlk1-Dio3 ncRNA cluster coordinates mitochondrial respiration and chromatin structure to maintain proper cell state.

The effect of tooth bleaching using violet LED (405-410 nm) on the properties of resin-based composites.

Photodiagnosis Photodyn Ther

December 2023

Department of Restorative Dentistry, University of São Paulo, School of Dentistry, Av. Prof. Lineu Prestes 2227, São Paulo, SP, 05508-000, Brazil.

Background: The objective of this study was to evaluate the effect of bleaching techniques, including or not the use of violet light (405-410 nm), on resin-based composites' color, surface roughness, nanohardness, and elastic modulus.

Methods: Ninety-six disk-shaped specimens (12 mm x 2 mm; n = 12) were prepared using Filtek Z350 XT (Z350) and IPS Empress Direct (ED) resin-based composites. After 24 h, specimens were stained in red wine for 28 days.

Oral Fluvoxamine With Inhaled Budesonide for Treatment of Early-Onset COVID-19 : A Randomized Platform Trial.

Ann Intern Med

May 2023

ViRx@Stanford, Stanford Biosecurity and Pandemic Preparedness Initiative, Stanford, California; Department of Health Research Methods, Evidence, and Impact, McMaster University, Hamilton, Ontario, Canada; Cytel, Vancouver, British Columbia, Canada; and Platform Life Sciences, Vancouver, British Columbia, Canada (E.J.M.).

Background: Previous trials have demonstrated the effects of fluvoxamine alone and inhaled budesonide alone for prevention of disease progression among outpatients with COVID-19.

Objective: To determine whether the combination of fluvoxamine and inhaled budesonide would increase treatment effects in a highly vaccinated population.

Design: Randomized, placebo-controlled, adaptive platform trial.

The aim of the present study was to investigate the impact of oral health literacy (OHL) on conceptions of care and behaviors related to COVID-19. The sample came from two preliminary cross-sectional studies that determined the level of OHL of parents/guardians of six-to-12-year-old children in two major Brazilian cities (Curitiba and Belo Horizonte). Functional OHL was measured using the Brazilian version of the Rapid Estimate of Adult Literacy in Dentistry (BREALD-30) and the Health Literacy Dental Scale (HeLD-14) for the evaluation of interactive OHL.

The Key Lnc (RNA)s in Cardiac and Skeletal Muscle Development, Regeneration, and Disease.

Non-coding RNAs (ncRNAs) play a key role in the regulation of transcriptional and epigenetic activity in mammalian cells. Comprehensive analysis of these ncRNAs has revealed sophisticated gene regulatory mechanisms which finely tune the proper gene output required for cellular homeostasis, proliferation, and differentiation. However, this elaborate circuitry has also made it vulnerable to perturbations that often result in disease.
