Here, we present a protocol to monitor and stimulate cardiac reprogramming via the disruption of Hdac4 condensates mediated by PC14-3-3 activation. We describe steps for inducing cardiac reprogramming in vitro, validating condensate formation ability of Hdac4, and observing Hdac4 condensates during reprogramming.
