Background: Agitation is a common and disabling symptom of Alzheimer's dementia (AD). Pharmacological treatments are recommended if agitation is not responsive to psychosocial intervention. Citalopram was effective in treating agitation in AD but was associated with cognitive and cardiac risks linked to its R- but not S-enantiomer.

Background: Benfotiamine, a prodrug of thiamine, raises blood levels by 50-100 times to achieve pharmacologic effects. It provides a novel therapeutic direction addressing a well-characterized brain tissue thiamine deficiency and related changes in glucose metabolism in AD. BenfoTeam is a seamless phase 2A-2B "proof of concept" (POC), double-blind, placebo-controlled RCT investigating tolerability, safety, and efficacy of benfotiamine, as a first-in-class small molecule treatment for early AD.

TREM2, a receptor expressed on microglia in the brain is critical to microglial function in health and in disease. Among microglia-associated AD risk genes, partial loss-of-function variants of TREM2 confer 2-3 fold increase in risk for developing AD, motivating efforts to identify pharmacological agonists targeting TREM2 as a therapeutic option.

Many digital therapeutic products predominantly focus on computerized cognitive training, lacking effective approaches to other crucial interventions. This study aimed to investigate the long term effects of multidimensional comprehensive cognitive intervention training program - Brain and Body Rehab Training (BBRT), which integrates multidomain cognitive training with physical-cognitive training and multidimensional lifestyle interventions on cognitive performance in participants with cognitive impairment after a 8-month follow-up.
