AI Article Synopsis

  • Merkel cell carcinoma (MCC) is a serious skin cancer with a 4.5% incidence of complete spontaneous regression (CRS), which suggests a better prognosis for those patients.
  • In a study of 112 MCC patients, the average age at diagnosis was 69, with tumors mainly found on the head and neck, and low immunosuppression rates among patients.
  • The presence of Merkel cell polyomavirus (MCPyV) was linked to regression, indicating that immune responses and tumor characteristics, along with further research, are needed to understand this phenomenon better.

Article Abstract

Merkel cell carcinoma (MCC) is an aggressive cutaneous neuroendocrine neoplasia. MCC complete spontaneous regression (CRS) has been linked to good prognosis. We conducted a retrospective review of a single-center cohort of MCC patients to analyze the characteristics of patients and tumors that exhibited regression. The calculated incidence of regressión was 4.5% (5/112). Median age at diagnosis was 69 years (RIQ 31), tumors were primarily located on the head and neck and patients had low rates of immunosuppression. After a median following-up of 4 years none of the patients showed recurrence or distant metastases. Merkel cell polyomavirus (MCPyV) positivity was associated with regression (P=0.045). Despite limited understanding of regression mechanisms, biopsy-induced immune responses linked with host and tumor factors such as age, immunosuppression or MCPyV positivity may play an important role. More research in this field is crucial for the comprehension of this rare phenomenon.

Sentinel Lymph Node Biopsy (SLNB) aims at identifying clinically occult nodal metastases. It is the standard staging procedure for patients with T1b to T4 primary cutaneous melanoma. Moreover, it is recommended whenever the risk of a positive SLNB is >5%, according to the National Comprehensive Cancer Network Melanoma guidelines.

Targeting of diseased cells is one of the most urgently needed prerequisites for a next generation of potent pharmaceuticals. Different approaches pursued fail mainly due to a lack of specific surface markers. Developing an RNA-based methodology, we can now ensure precise cell targeting combined with selective expression of effector proteins for therapy, diagnostics or cell steering.

In this study, an advanced nanofiber breast cancer model was developed and systematically characterized including physico-chemical, cell-biological and biophysical parameters. Using electrospinning, the architecture of tumor-associated collagen signatures (TACS5 and TACS6) was mimicked. By employing a rotating cylinder or static plate collector set-up, aligned fibers (TACS5-like structures) and randomly orientated fibers (TACS6-like structures) fibers were produced, respectively.

Objective: The lack of consensus on the benefits and harms of standard therapies, including surgery (SRx), radiotherapy (RTx), chemotherapy (CTx), and their combinations among early-stage MCC, prompted this study.

Methods: A systematic review and meta-analysis of randomized and non-randomized studies published between January 01, 1972, and January 31, 2023, and having overall survival (OS), local recurrence (LR), regional recurrence (RR), disease-specific survival (DSS), and/or disease-free survival (DFS) as outcomes was conducted using the Cochrane Central Register of Controlled Trials (CENTRAL), PubMed (NCBI), Scopus (ELSEVIER), and Web of Science (CLAVIRATE) databases. Hazard ratios (HRs) and their variances were pooled using the inverse variance heterogeneity model.

Capicua transcriptional repressor (CIC)-rearranged sarcoma (CRS) is a rare and recently described tumor that most commonly affects patients between 15 and 30 years of age. It is an undifferentiated round cell malignancy, with a disease defining CIC fusion, with double homeobox 4 (DUX4) being the most common partner. Here, we report a 77-year-old woman who presented with a cutaneous thigh mass with a clinical morphology suggesting Merkel cell carcinoma.
