Enhancing farmed fish welfare: Evaluating the effectiveness of plant-based stress mitigating agents as sedatives in sea bass (Dicentrarchus labrax) following intraperitoneal vaccination.

The effectiveness of vaccines may be compromised by the stress response induced by intraperitoneal/intramuscular (IP/IM) vaccination due to an intimate interaction between the neuroendocrine and immune systems. Essential oils (EOs), known for their antibacterial, antioxidant, and sedative properties, are potential candidates to mitigate this stress response. This study investigates the short-term sedative effect of two essential oil-based products, FishEase-C (FEC) and FishEase-L (FEL) in sea bass prior to intraperitoneal vaccination (IP). Physiological stress indicators (plasma cortisol, glucose, lactate) increased 1-h post-vaccination (hpv) as expected. Cortisol remained elevated up to 24 hpv in the N.P. vaccine and FEC + vaccine groups but decreased with FEL treatment (FEL + vaccine group). However, FEC at the tested concentration appeared to induce stress. The transcription of stress (gr, hsp70, cox2), immune (il1β, il6, tnfα), and antioxidant (gpx, sod, catalase) genes confirmed the vaccination-induced stress response, with mc2r transcription indicating increased cortisol production in vaccinated groups (N.P. vaccine and FEC + vaccine). FEL reduced stress at both physiological (e.g., cortisol) and transcriptional levels (e.g., hsp70, cox2 and il6) at either 1 or 24 hpv. It is worth noting that, from an inflammatory perspective, there was a big difference between tissues in terms of magnitude and pattern (treatment and time effects). The brain was more resistant to inflammation, while the head kidney and spleen showed heightened il1β expression (860-2100-fold). These findings support the use of FEL as a sedative before IP/IM vaccination in sea bass.

The potential risk posed by infectious agents (IAs) associated with netpen aquaculture to wild fishes is determined based on the "release" of IAs from netpens into the environment, the "exposure" of the wild fish to those released agents, and the "consequence" for wild fish experiencing infection by those agents. Information available to characterize these three factors is often lacking, and the occurrence of transmission from aquaculture to wild fish as well as potential consequences of such transmission are difficult to observe. The present study was to investigate the effect of the astaxanthin high internal phase Pickering emulsion (H-AXT) on DEHP-induced liver lipid metabolism disorder and to demonstrate its possible protective mechanism. We have developed an antioxidant activity emulsion system to deliver astaxanthin into the liver to maximize its ability to protect the liver. In vitro, H-AXT intervention inhibited oxidative stress restored the level of mitochondrial membrane potential to 90 % of that of normal LO2 cells, and alleviated the imbalance of energy metabolism by protecting mitochondrial structure and function.

Ammonia, a ubiquitous contaminant in aquatic ecosystems, poses multifaceted threats to fish species at elevated concentrations. In order to investigate the toxic effects of chronic ammonia stress on the liver of juvenile , the present experiment was conducted to investigate the differences in changes in liver tissue structure, enzyme activities, and metabolomes after 28 days of ammonia exposure (0, 4, 8, and 16 mg/L). The findings revealed that ammonia exposure induced significant oxidative stress in the liver, manifesting in decreased activities of antioxidant enzymes SOD and GSH-Px, elevated levels of GSH, GST, and MDA, and heightened activities of immune enzymes LZM, ALP, and ACP.

Accurate methods for assessing food freshness through colorimetric pH response play a critical role in determining food spoilage and ensuring food quality standards. This study introduces a novel unlabeled DNA sequence, poly-dA, designed to exploit the colorimetric properties of both the single strand and the fold-back A-motif structure in conjunction with gold nanoparticles (AuNPs) under varying pH conditions. When exposed to storage temperatures of 4 °C and 25 °C, the color variations in the AuNP solution, influenced by pH level changes in mutton and sea bass samples' different storage periods, are easily discernible to the naked eye within a minute.
