R2 retrotransposons are model site-specific eukaryotic non-LTR retrotransposons that copy-and-paste into gene loci encoding ribosomal RNAs. Recently we demonstrated that avian A-clade R2 proteins achieve efficient and precise insertion of transgenes into their native safe-harbor loci in human cells. The features of A-clade R2 proteins that support gene insertion are not characterized. Here, we report high resolution cryo-electron microscopy structures of two vertebrate A-clade R2 proteins, avian and testudine, at the initiation of target-primed reverse transcription and one structure after cDNA synthesis and second strand nicking. Using biochemical and cellular assays we discover the basis for high selectivity of template use and unique roles for each of the expanded A-clade zinc-finger domains in nucleic acid recognition. Reverse transcriptase active site architecture is reinforced by an unanticipated insertion motif in vertebrate A-clade R2 proteins. Our work brings first insights to A-clade R2 protein structure during gene insertion and enables further improvement and adaptation of R2-based systems for precise transgene insertion.

Little is known about the insect viruses in wheat sawfly, Dolerus tritici, which is an important agricultural insect feeding on wheat leaves. Here, we used RNA sequencing to identify a novel single positive-strand RNA virus from the larvae of wheat sawfly collected in northern China and then determined its complete genome sequence by rapid amplification of cDNA ends. The complete genome is 9,594 nt in length, including a polyA tail at its 3' terminus, and it is predicted to encode a 326.

A new genus and species within the family Polymorphidae Meyer, 1931 were erected to accommodate cystacanths recovered from the mesentery of individuals from a non-native population of the red swamp crayfish Procambarus clarkii (Girard), collected from South Carolina (USA). Morphological characteristics of the specimens collected included in both sexes a spindle-shaped body with a slender hindtrunk, two fields of markedly different sized spines on the foretrunk, the presence of a middle row of smaller scythe-shaped hooks on a proboscis armed with 19-20 longitudinal rows of 14-16 hooks; and in males, six cement glands, absence of genital spines, and a digitiform and spinose bursa (observed inverted). Sequencing portions of both the mitochondrial cytochrome c oxidase I (COI) and large subunit ribosomal RNA genes was completed, followed by phylogenetic analysis of a concatenated alignment.

During parasitological surveys of marine fishes and zooplankton in localities of the Northwestern Pacific coast of Mexico, 28 Gafftopsail pompano (Trachinotus rhodopus Gill) and 50 mysid crustaceans (Metamysidopsis frankfiersi Hendrickx & Hernández-Payán) we identified to be infected with adults and cystacanths, respectively of an acanthocephalan morphologically corresponding to the genus Rhadinorhynchus Lühe, 1911. DNA sequences of the small (SSU) and large (LSU) subunits of ribosomal DNA, and cytochrome c oxidase subunit 1 (cox 1) from mitochondrial DNA were obtained. Phylogenetic analyses revealed that the newly sequenced individuals in a clade with Rhadinorhynchus sp.
