Cohesin complex is essential for cell division and regulating cell type-specific gene expression programs. Mutations in genes encoding the cohesin subunits are associated with hematological malignancies, preleukemia, and clonal hematopoiesis of indeterminate potential. In this study, we examined how cohesin mutation impacts hematopoiesis using adult zebrafish that carry heterozygous germline nonsense mutation in the cohesin subunit, () that is orthologous to human . Single-cell RNA sequencing analyses showed that adult zebrafish harboring mutation exhibit significant transcriptional dysregulation within the whole kidney marrow and have altered erythroid and granulocyte output. Erythroid progenitors were expanded in and erythroid differentiation was altered. The expression profile of several erythroid genes, including , was dysregulated in erythroid cells. Mature granulocyte population declined in , and the transcriptional program of granulocytes was impaired but granulocytic maturation was maintained. Granulocytes from showed upregulation of stress hematopoiesis factor, . These findings show that normal is required to maintain steady erythropoiesis and granulopoiesis in the adult zebrafish marrow. Mutations in cohesin subunit genes are early events in leukemogenesis. This study characterizes the hematopoietic compartment of adult zebrafish that carry germline heterozygous mutation in cohesin subunit, . Our results show that despite normal appearance, mutant adult zebrafish exhibit transcriptional dysregulation and altered erythroid and granulocyte output. No obvious morphological dysplasia was observed in the mutant hematopoietic cells. These results suggest that mutation can cause underlying hematopoietic disturbances.

Microcystin-leucine-arginine (MC-LR) has been shown to induce neuroinflammation and disrupt neurotransmitter system. However, little is known about the mechanism of toxicity. In this study, male adult zebrafish (Danio rerio) were exposed to MC-LR at concentrations of 0, 0.

Ex vivo electroretinography (ERG) provides insight into the health and functionality of retinal cells, the integrity of phototransduction, and the visual cycle and allows for the direct application of pharmaceuticals to the retinal tissues. Here, we present a protocol for performing ex vivo ERG on adult zebrafish. We describe steps for zebrafish tissue dissection, mounting tissues, and assembling and connecting cassettes.

Cannabis is one of the most widely used drugs, and yet an understanding of its impact on the human brain and body is inconclusive. Medicinal and recreational use of cannabis has increased in the last decade with a concomitant increase in use by pregnant women. The major psychoactive compound in cannabis, Δ-tetrahydrocannabinol (THC), exists in different isomers, with the (-) trans isomer most common.

Heritable fragile bone disorders (FBDs), ranging from multifactorial to rare monogenic conditions, are characterized by an elevated fracture risk. Validating causative genes and understanding their mechanisms remain challenging. We assessed a semi-high throughput zebrafish screening platform for rapid in vivo functional testing of candidate FBD genes.

Stress plays a key role in mental, neurological, endocrine, and immune disorders. The zebrafish (Danio rerio) is rapidly gaining popularity as s model organism in stress physiology and neuroscience research. Although the leopard (leo) fish are a common outbred zebrafish strain, their behavioral phenotypes and stress responses remain poorly characterized.
