Background: Alzheimer's disease (AD) is a progressive neurodegenerative disease associated with neuroinflammation and heightened production of reactive oxygen species (ROS) in the brain from overactive NADPH Oxidase 2 (NOX2). The current study examines whether administration of a novel, brain-penetrant NOX2 inhibitor (CPP11G & CPP11H) reduces amyloid plaque load and improves AD-associated vascular dysfunction in a male APP-PS1 mouse model of AD.

Method: Intraperitoneal injections of CPP11G (n = 1) or CPP11H (n = 2) three times per week began at 9-10 months of age in the treatment APP-PS1 group (15 mg/kg).

Memory and Aging Center, UCSF Weill Institute for Neurosciences, University of California, San Francisco, San Francisco, CA, USA.

Background: Aging associates with decreased functional connectivity between brain regions linked to musical rhythm perception. Producing rhythmic music may result in strengthened functional connectivity of these regions, but more evidence is needed to support intervention design. Currently, few studies directly contrast younger and older adults' rhythmic music performance to understand brain-behavior relationships.

Background: Dementia, including its younger-onset variant, increasingly challenges diverse workplaces, affecting both employees living with dementia and their employers. With a growing global trend of individuals extending their working years, it becomes crucial to understand the specific challenges and implications for those with dementia within various employment contexts. This review delves into the cognitive, interpersonal, and organizational challenges faced by employees living with dementia across a broad range of workplaces, reflecting the evolving dynamics of today's work environment.

Background: Caregiver burden, or the impact of caregiving, commonly occurs in caregivers of persons with dementia (PWD); however, prior research focused on caregiver burden in this population has not considered the impact of dysphagia. Therefore, the purpose of this study was to measure dysphagia-specific burden in caregivers of PWD and examine its relationship to general caregiver burden, as well as the PWD's current diet level and dementia severity.

Method: Data were collected from PWD-caregiver dyads participating in a prospective, dysphagia-focused clinical trial at the initial study visit.

Alzheimers Dement

December 2024

University of Kentucky College of Medicine, Sanders-Brown Center on Aging, Lexington, KY, USA.

Background: We currently lack in the dementia field accurate, noninvasive, quick, and affordable screening tools for brain dysfunctions associated with early subtle risk of mild cognitive impairment (MCI). Our Kentucky aging cohort demonstrates that asymptomatic older individuals with MCI-like frontal memory-related brainwave patterns convert to MCI within a short 5-year period, as opposed to individuals with NC-like patterns (1) that remain normal 10 years later (2). Astrocyte reactivity influences amyloid-ß effects on tau pathology in preclinical Alzheimer's disease (3).
