Article Abstract

Modulating RNA decay is a powerful tool to investigate RNA degradation dynamics. Here, we describe a protocol to inducibly recruit protein factors to regulate target RNA metabolism, called Rapid Inducible Decay of RNA (RIDR). RIDR induces fast and synchronous decay of target mRNAs within minutes and enables direct visualization of mRNA decay dynamics and subcellular kinetics in living cells. Here, we provide detailed procedures to make stable cell lines, conduct fixed- and live-cell measurements, and perform data analysis. We discuss the potential pitfalls and make RIDR applicable to a general biology lab.

Transgenerational plasticity (TGP) has largely focused on how parental exposure to ecological conditions shapes the phenotypes of future generations. However, organisms acquire information about their ecological environment via social learning, which can also shape TGP in profound ways. We demonstrate that non-parents alter how parents detect and respond to environmental cues in ways that spillover to affect offspring, non-parents influence offspring even without direct physical interactions, and parental cues received by offspring can alter the phenotypes of other juveniles.

The plasticity of plant cells underlies their wide capacity to regenerate, with increasing evidence in plants and animals implicating cell-cycle dynamics in cellular reprogramming. To investigate the cell cycle during cellular reprogramming, we developed a comprehensive set of cell-cycle-phase markers in the Arabidopsis root. Using single-cell RNA sequencing profiles and live imaging during regeneration, we found that a subset of cells near an ablation injury dramatically increases division rate by truncating G1 phase.
