AI Article Synopsis

  • Malaria continues to be a leading cause of death globally, and existing treatments are becoming less effective due to drug resistance, prompting the need for new drug discovery methods.
  • The study integrates machine learning with quantitative structure-activity relationship (QSAR) techniques to create a model for identifying potential new malaria drug candidates based on a polynomial equation.
  • A web-based software tool, PlasmoQSAR, was developed to help researchers estimate the effectiveness of new molecules against malaria by calculating their half maximal effective concentration (EC).

Article Abstract

Despite new therapies against malaria, this disease remains one of the main causes of death affecting humanity. The phenomenon of resistance has caused concern as the drugs no longer have the same efficacy, forcing scientific research to develop new methods that prospect new molecules. With the advancement of artificial intelligence, it becomes possible to apply machine learning (ML) techniques in the discovery and evaluation of new molecules, by employing the quantitative structure-activity relationship (QSAR), a classic method that uses regressions to create a model that allows identifying and evaluating new drug candidates. This work combined QSAR with ML and developed a supervised model that modeled a fourth degree polynomial equation capable of identifying new drug candidates derived from the triclosan compound-a classic inhibitor of growth, the cause of severe malaria. The model produces an greater than 80% for training and concurrent testing, as well as a correlation index greater than 80% between the calculated and experimental pEC (negative logarithm of half maximal effective concentration) data. In addition, a web software (PlasmoQSAR) was created that allows researchers to calculate the EC (half maximal effective concentration) of new molecules using the developed analytical method.

Background: Accurate classification of host phenotypes from microbiome data is crucial for advancing microbiome-based therapies, with machine learning offering effective solutions. However, the complexity of the gut microbiome, data sparsity, compositionality, and population-specificity present significant challenges. Microbiome data transformations can alleviate some of the aforementioned challenges, but their usage in machine learning tasks has largely been unexplored.

Department of Hematology, The First Affiliated Hospital of Xiamen University and Institute of Hematology, School of Medicine, Xiamen University, Xiamen, 361003, P.R. China.

Background: Disease progression within 24 months (POD24) significantly impacts overall survival (OS) in patients with follicular lymphoma (FL). This study aimed to develop a robust predictive model, FLIPI-C, using a machine learning approach to identify FL patients at high risk of POD24.

Methods: A cohort of 1,938 FL patients (FL1-3a) from seventeen centers nationwide in China was randomly divided into training and internal validation sets (2:1 ratio).

School of Medicine, Chinese PLA General Hospital, Nankai University, Beijing, China.

Background: Glioblastoma multiforme (GBM) exhibits a cellular hierarchy with a subpopulation of stem-like cells known as glioblastoma stem cells (GSCs) that drive tumor growth and contribute to treatment resistance. NAD(H) emerges as a crucial factor influencing GSC maintenance through its involvement in diverse biological processes, including mitochondrial fitness and DNA damage repair. However, how GSCs leverage metabolic adaptation to obtain survival advantage remains elusive.
