Chromatin immunoprecipitation (ChIP) assay is a standard method to examine protein-DNA interactions in vivo. Here, I describe a basic procedure for ChIP assay in rice shoot apices using an antibody against the homeodomain transcription factor Oryza sativa homeobox1 (OSH1). I also introduce the tips to overcome several obstacles when performing this assay for the first time and potential pitfalls that could impact the result.

The epigenetic state of chromatin, gene activity and chromosomal positions are interrelated in plants. In Arabidopsis thaliana, chromosome arms are DNA-hypomethylated and enriched with the euchromatin-specific histone mark H3K4me3, while pericentromeric regions are DNA-hypermethylated and enriched with the heterochromatin-specific mark H3K9me2. Background: Non-thyroidal illness syndrome is commonly observed in critically ill patients, characterized by the inactivation of systemic thyroid hormones (TH), which aggravates metabolic dysfunction. Recent evidence indicates that enhanced TH inactivation is mediated by the reactivation of type 3 deiodinase (Dio3) at the tissue level, culminating in a perturbed local metabolic equilibrium. This study assessed whether targeted inhibition of Dio3 can maintain tissue metabolic homeostasis under septic conditions and explored the mechanism behind Dio3 reactivation.
