is an edible ectomycorrhizal mushroom that forms a symbiotic association with Pinaceae trees by constructing a large extraradical mycelial area (called a shiro) in the soil. The detection of this fungal mycelium in the soil is crucial for estimating the success of outplanted mycorrhizal seedlings inoculated with under experimental conditions. Although several -specific DNA markers have been reported for efficient detection in the field, no comparative study has been conducted to assess their effectiveness. In the present study, we targeted the nuclear ribosomal DNA intergenic spacer 2 (IGS2) region for the detection of . The newly designed TmSP-I-2F/TmSP-I-2R primer pair, which targets a partial IGS2 sequence (543 bp), effectively detected from pine root and soil samples via PCR assay, outperforming other -specific primers. In combination with a PCR system targeting LTR DNA markers that were previously developed, a PCR system with the TmSP-I-2F/TmSP-I-2R primer pair set can expedite investigations of the dynamics of genets in mycorrhizas and shiro.

Black-goggled tanagers Trichothraupis melanops (Vieillot, 1818) are passerine birds commonly observed in the Brazilian Atlantic Forest, Argentina and Paraguay. Tanagers are among the passerines with the highest prevalence and density of coccidian parasites, mainly due to their frugivorous feeding habits that favor fecal-oral transmission. In this context, the current study identifies a new species of Isospora Schneider, 1881 parasitizing black-goggled tanagers captured in the Itatiaia National Park, a protected area with a high degree of vulnerability in Southeastern Brazil.

Peucedanum japonicum (PJ), a member of the Apiaceae family, is widely distributed and cultivated in East Asian countries for edible and functional foods. In this study, we compared the plastid genomes (plastomes) and 45S nuclear ribosomal DNA (45S nrDNA) simultaneously from 10 PJ collections. Plastome-based phylogenetic analysis showed that the PJ accessions were monophyletic within the genus Peucedanum.

Pseudouridine (Ψ) is a post-transcriptional modifier of RNA, often referred to as the 'fifth nucleotide' owing to its regulatory role in various biological functions as well as because of its significant involvement in the pathogenesis of human cancer. In recent years, research has revealed various Ψ modifications in different RNA types, including messenger RNA, transfer RNA, ribosomal RNA, small nuclear RNA, and long noncoding RNA. Pseudouridylation can significantly alter RNA structure and thermodynamic stability, as the Ψ-adenine (A) base pair is more stable than the typical uridine (U)-A base pair is due to its structural similarity to adenine.

Multicopy nuclear ribosomal DNA (rDNA) genes have been used as markers for fungal identification for three decades. The rDNA sequences in a genome are thought to be homogeneous due to concerted evolution. However, intragenomic variation of rDNA sequences has recently been observed in many fungi, which may make fungal identification and species abundance estimation based on these loci problematic.

In plants, cellular function is orchestrated by three distinct genomes located within the nucleus, mitochondrion, and plastid. These genomes are interdependent, requiring tightly coordinated maintenance and expression. Plastids host several multisubunit protein complexes encoded by both the plastid and nuclear genomes.
