Aim: The aim of the study was to evaluate the effect of thiolation of lipid-based nanoparticles (LNPs) on cellular uptake of nucleic acids.

Methods: A thiolated surfactant was synthesized by binding palmitic acid covalently to cysteine. Green fluorescent protein (GFP) encoding plasmid DNA (pDNA) was used as model nucleic acid and incorporated via hydrophobic ion-pairing with a cationic cholesterol derivate (DcCholesterol) in LNPs that were prepared by solvent injection method using the thiolated surfactant for surface decoration. LNPs were characterized regarding size, polydispersity index, zeta potential and stability in biorelevant media. The endosomal escape properties of LNPs were evaluated by erythrocytes interaction studies. Cell viability and transfection efficiency on HEK293 cells were investigated.

Results: The structure of the thiolated surfactant was confirmed by H NMR and FT-IR. LNPs containing the nucleic acid-DcCholesterol complex with and without the thiolated surfactant were developed and displayed sizes in the range from 173 nm to 233 nm with a narrow size distribution (PDI < 0.3) and a negative zeta potential. LNPs showed no significant increase in size after 4 h of incubation in artificial body fluids. Erythrocytes interaction studies revealed enhanced endosomal escape properties for thiolated LNPs compared to non-thiolated LNPs. LNPs showed in concentrations lower than 0.74 mg/mL cell viability ≥ 80 %. Transfection studies on HEK cells with thiolated LNPs compared to non-thiolated LNP showed a 4.6-fold higher expression of GFP.

Conclusion: Surface thiolation of LNPs represents a promising tool for enhancing intracellular nucleic acid delivery of LNPs.

The semisynthetic derivatization of natural products is crucial for their continued development as antibiotics. While commercial pleuromutilin derivatives depend on amines for solubility, we demonstrate the high activity and solubility of oligoethylene glycol-substituted pleuromutilins achieved via a one-pot deprotection/attachment approach using thiolates protected as thioesters. The bifunctional linker synthesis is versatile and can be broadly applied to other chemistries.

Effect of End-Tethered Methoxy-PEO Chain Density on Uremic Toxin Adsorption.

ACS Appl Bio Mater

January 2025

Department of Chemical and Materials Engineering, University of Alberta, Edmonton, Alberta T6G 1H9, Canada.

In 2023, around 850 million people globally were affected by chronic kidney disease, which leads to the retention of uremic toxins and excess fluid in the blood. This study examines the adsorption of these toxins to poly(ethylene oxide) (PEO) films, known for their low-fouling properties. The gold surfaces were treated with 5 mM end-thiolated methoxy-terminated PEO (-PEO) and analyzed using dynamic contact angle measurements, X-ray photoelectron spectroscopy, and spectroscopic ellipsometry to confirm the PEO film's presence and determine chain density.

December 2024

Department of Chemistry, Institute for Molecular Science and Fusion Technology, Kangwon National University, Chuncheon 24341, Korea.

Accurate detection of glucose and insulin is crucial for early diagnosis, classification, and timely prevention of diabetes. In this study, we present a novel surface-enhanced Raman scattering (SERS) aptasensor for glucose and insulin detection. The SERS aptasensor is composed of gold bipyramidal nanoparticles (Au BPs), SH-aptamer-methylene blue (MB), and thiolated polyethylene glycol (SH-PEG).

Article Synopsis
  • - Organometallic oxidative addition complexes (OACs) are gaining attention as effective reagents for the selective modification of biomolecules by altering ligand and aryl properties to control reaction kinetics and regioselectivity.
  • - Researchers explored the use of bidentate Au(III) OACs with bulky and electron-deficient aryl substrates to successfully achieve selective -arylation using computational and experimental methods.
  • - The study successfully developed a protein-polymer OAC that performed rapid -arylation with designed ankyrin repeat proteins (DARPins) and various biologically relevant small molecules, paving the way for constructing complex biomolecular conjugates.
