Here, we optimized the active units of polysaccharides and investigated the conformational relationship between the polysaccharides and alcoholic liver disease (ALD) at the molecular level. We used data mining to screen polysaccharide structural parameters for ALD (PSP-ALD). Most ALD-resistant polysaccharides against ALD comprised glucose (Glc), mannose (Man), galactose (Gal), arabinose (Ara), and rhamnose (Rha). Additionally, (1 → 6)-, (1 → 3)-, and (1 → 4)- glycosidic linkages were mainly contained. Polysaccharides against ALD have a wide molecular weight distribution (2.1 × 10 Da - 9.6 × 10 Da). Based on the PSP-ALD analysis, six commercially available oligosaccharides were selected and their structures were built. After molecular docking, the binding affinities between stachyose and the key ALD targets were stronger, indicating that stachyose may be a polysaccharide-active unit against ALD (PAU-ALD). Furthermore, histological examination of liver tissue combined with serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and triglycerides (TG) showed that stachyose had a significant protective effect against ALD in mice. In summary, we optimized a PAU-ALD and developed a method for studying the structure-activity relationship between polysaccharides and ALD at the molecular level, which provides a new research direction for the development and utilization of polysaccharides and their clinical applications in ALD.

Background & Aims: Hepatic encephalopathy (HE), one of the most serious prognostic factors for mortality in alcohol-related cirrhosis (ALD cirrhosis), is not recorded in Danish healthcare registries. However, treatment of HE with lactulose, the universal first-line treatment, can be identified through data on filled prescriptions. This study aimed to investigate if lactulose can be used as a surrogate marker of HE.

The efficient extraction and purification of active components from Eucommia ulmoides Oliver (EUO) are crucial for their utilization. The structure and properties of the prepared EUO leaf polysaccharides (ELPs) and extractum (ELE) were comprehensively characterized in this study, and the intervention mechanism of the EUO polysaccharides and extractum in alcoholic liver disease (ALD) were investigated. The yield of EUO extractum was 24.

Background: Liuweizhiji Gegen-Sangshen oral liquid (LGS), as a Chinese medicinal preparation, is developed from a Traditional Chinese medicinal formula consisting of six Chinese medicinal herbs, including Puerariae lobatae radix, Hoveniae semen, Imperatae rhizoma, Crataegi fructus, Mori fructus and Canarli fructus, and has been extensively utilized in the prevention and treatment of alcoholic liver disease (ALD) clinically. Previous study has demonstrated that LGS dose-dependently mitigated ALD in rat models. However, whether and how the main characteristic constituents of LGS (the flavonoid and polysaccharide fractions, LGSF and LGSP) contribute to the anti-ALD effect remains unclear.

This study aimed to ascertain the potential benefits of green radish polysaccharide (GRP) in treating alcoholic liver disease (ALD) in mice and explore its mechanism of action. Using biochemical analysis, high-throughput sequencing of gut microbiota, and gas chromatography-mass spectrometry to measure short-chain fatty acids (SCFAs) in feces, we found that GRP intervention significantly improved lipid metabolism and hepatic function in mice subjected to excessive alcohol intake. The GRP intervention reduced malondialdehyde levels by 66% and increased total superoxide dismutase levels by 22%, thereby mitigating alcohol-induced oxidative stress.
