  • L. Webb ex Prantl is widely used in traditional medicine but lacks an effective genetic transformation system for research and application.
  • This study presents a new, efficient method for genetic transformation that involves dipping inflorescences in a specific solution for 45 seconds, leading to successful plant transformations.
  • The method also incorporates CRISPR/Cas9 for targeted genome editing, enabling the creation of mutant plants with a specific trait in just 2.5 months.

Article Abstract

L. Webb ex Prantl is used in traditional medicine globally. However, the lack of an efficient and reliable genetic transformation system has seriously limited the investigation of gene function and further utilization of . In this study, a highly efficient, time-saving, and cost-effective -mediated genetic transformation system has been developed in . In this method, the transformation was accomplished by simply dipping developing inflorescences for 45 s into an suspension (OD = 0.6) containing 5% sucrose and 0.03% (/) Silwet L-77. Treated plants were allowed to set seeds which were then plated on a selective medium with hygromycin B (HygB) to screen transformants. Additionally, the CRISPR/Cas9 genomic editing system was validated by targeting phytoene desaturase () gene using this floral dip method, and mutant plants with the expected albino phenotype could be obtained in 2.5 months. This genetic transformation and targeted editing system will be a valuable tool for routine investigation of gene function and further exploitation in .

Agrobacterium-mediated gene transformation method is a vital molecular biology technique employed to develop transgenic plants. Plants are genetically engineered to develop disease-free varieties, knock out unsettling traits for crop improvement, or incorporate an antigenic protein to make the plant a green factory for edible vaccines. The method's robustness was validated through successful transformations, demonstrating its effectiveness as a standard approach for researchers working in plant biotechnology.

Frontal Fibrosing Alopecia Part II: Etiopathogenesis and Management.

J Am Acad Dermatol

January 2025

Dr. Phillip Frost Department of Dermatology and Cutaneous Surgery, University of Miami Miller School of Medicine, Miami, FL.

Frontal Fibrosing Alopecia (FFA) poses a distinct dermatological challenge with epithelial-mesenchymal transition (EMT) at its core, driving follicular cell transformation and fibrotic changes. Genetic studies highlight significant associations, while environmental triggers, such as implicated cosmetic products (sunblock, personal hair care products, and moisturizers), introduce complexity. Managing FFA proves daunting due to its chronic and unpredictable nature.

The plastid-encoded RNA polymerase (PEP) plays an essential role in the transcription of the chloroplast genome. Here, we present a strategy to purify the transcriptionally active protein complex from transplastomic tobacco (Nicotiana tabacum) lines in which one of the PEP core subunits is fused to an epitope tag. We describe experimental procedures for designing transformation constructs for PEP purification, selection, and analysis of transplastomic tobacco plants.

The Role of Chronic Inflammation in Pediatric Cancer.

Cancers (Basel)

January 2025

Division of Hematology Oncology, Akron Children's Hospital, One Perkins Square, Akron, OH 44308, USA.

Inflammation plays a crucial role in wound healing and the host immune response following pathogenic invasion. However, unresolved chronic inflammation can result in tissue fibrosis and genetic alterations that contribute to the pathogenesis of human diseases such as cancer. Recent scientific advancements exploring the underlying mechanisms of malignant cellular transformations and cancer progression have exposed significant disparities between pediatric and adult-onset cancers.
