The solute carrier 17 (SLC17) family contains anion transporters that accumulate neurotransmitters in secretory vesicles, remove carboxylated monosaccharides from lysosomes, or extrude organic anions from the kidneys and liver. We combined classical molecular dynamics simulations, Markov state modeling and hybrid first principles quantum mechanical/classical mechanical (QM/MM) simulations with experimental approaches to describe the transport mechanisms of a model bacterial protein, the D-galactonate transporter DgoT, at atomic resolution. We found that protonation of D46 and E133 precedes galactonate binding and that substrate binding induces closure of the extracellular gate, with the conserved R47 coupling substrate binding to transmembrane helix movement. After isomerization to an inward-facing conformation, deprotonation of E133 and subsequent proton transfer from D46 to E133 opens the intracellular gate and permits galactonate dissociation either in its unprotonated form or after proton transfer from E133. After release of the second proton, apo DgoT returns to the outward-facing conformation. Our results provide a framework to understand how various SLC17 transport functions with distinct transport stoichiometries can be attained through subtle variations in proton and substrate binding/unbinding.

Secretory diarrhea, a major global health concern, particularly among young children, is often characterized by excessive chloride secretion through the cystic fibrosis transmembrane conductance regulator (CFTR) channel. Nornidulin, a fungus-derived natural product from Aspergillus unguis, has previously been shown to inhibit cAMP-induced Cl- secretion in T84 cells (human intestinal cell lines). However, the cellular mechanism of nornidulin in inhibiting cAMP-induced Cl- secretion and its anti-secretory efficacy is still unknown especially in a human colonoid model, a preclinical model recapitulating intestinal physiology in humans.

Manganese-based (Mn-based) layered oxides have emerged as competitive cathode materials for sodium-ion batteries (SIBs), primarily due to their high energy density, cost-effectiveness, and potential for mass production. In this study, the rhombohedral crystalline pure phase BiFeO (BFO) of irregularly shaped spherical particles of ≈100 nm and energy bandgap of ≈2.31 eV are synthesized by sol-gel auto-combustion method and explored as electrode material for photo-assisted supercapacitor. The electronic structure studies revealed that the coexistence of heterovalent Bi and Fe elements accelerated the electrochemical redox kinetics and photo-assisted charge storage properties.

Highly concentrated salt solutions are promising electrolytes for battery applications due to their low flammability, their high thermal stability, and their good compatibility with electrode materials. Understanding transport processes in highly concentrated electrolytes is a challenging task, since strong ion-ion and ion-solvent interactions lead to highly correlated movements on the microscopic scale. Here, we use an experimental overdetermination method to obtain accurate Onsager transport coefficients for concentrated binary electrolytes composed of either sulfolane (SL) or dimethyl carbonate (DMC) as solvent and either LiTFSI or LiFSI as salt.
