Herein, we present a prominent metal-free C-N cross-coupling platform that enables access to carbamoyl- and ketoazides from isocyanides or silyl enol ethers and trimethylsilyl azide (TMSN) with an aid of iodine(III) promoter. This offers a rapid route to a diverse set of synthetically valuable azide decorated fragments with excellent substrate scope and good to excellent yields. The disclosed platform exemplifies the use of TMSN for incorporation of the azide fragment without the loss of N.

1,4-Dibenzodiazepines, an important component of nitrogen-containing heterocycles, are widely present in drugs. Herein, we developed a photochemical radical cascade cyclization reaction of isocyanides with α-carbonyl bromides under mild conditions. A sequence of 11-alkyl-substituted 1,4-dibenzodiazepines were produced in 53%-85% yields, demonstrating excellent tolerance towards various functional groups.

This study investigates the potential of boron trifluoride etherate (BF·OEt) to trigger unprecedented reactions of 2-oxoaldehydes with nitriles and amides/sulphonamides. In contrast to the mechanism in conventional reactions, the α-carbonyl group in 2-oxoaldehydes induces a cyclization pathway to be followed when reacting with nitriles, yielding 4-amidooxazoles. Additionally, reactions with weak nucleophiles produce β-keto amides/sulphonamides.

Reductive amination is one of the most synthetically direct routes to access chiral amines. Several Imine Reductases (IREDs) have been discovered to catalyze reductive amination (Reductive Aminases or RedAms), yet they are dependent on the expensive phosphorylated nicotinamide adenine dinucleotide cofactor NADPH and usually more active at basic pH. Here, we describe the discovery and synthetic potential of an IRED from (RedAm) that catalyzes reductive amination between a series of medium to large carbonyl and amine compounds with conversions of up to >99% and 99% enantiomeric excess at neutral pH.
