Laue-DIALS: Open-source software for polychromatic x-ray diffraction data.

Most x-ray sources are inherently polychromatic. Polychromatic ("pink") x-rays provide an efficient way to conduct diffraction experiments as many more photons can be used and large regions of reciprocal space can be probed without sample rotation during exposure-ideal conditions for time-resolved applications. Analysis of such data is complicated, however, causing most x-ray facilities to discard >99% of x-ray photons to obtain monochromatic data. Key challenges in analyzing polychromatic diffraction data include lattice searching, indexing and wavelength assignment, correction of measured intensities for wavelength-dependent effects, and deconvolution of harmonics. We recently described an algorithm, Careless, that can perform harmonic deconvolution and correct measured intensities for variation in wavelength when presented with integrated diffraction intensities and assigned wavelengths. Here, we present Laue-DIALS, an open-source software pipeline that indexes and integrates polychromatic diffraction data. Laue-DIALS is based on the dxtbx toolbox, which supports the DIALS software commonly used to process monochromatic data. As such, Laue-DIALS provides many of the same advantages: an open-source, modular, and extensible architecture, providing a robust basis for future development. We present benchmark results showing that Laue-DIALS, together with Careless, provides a suitable approach to the analysis of polychromatic diffraction data, including for time-resolved applications.

Nidustrin A (1), the first cysteine-retained emestrin featuring a unique sulfur-containing 18-membered macrocyclic lactone, along with four biogenetically related compounds (2-5), and one known analogue secoemestrin C (6), were isolated from the large-scale culture of Aspergillus nidulans, an endophytic fungus derived from the Whitmania pigra. Compounds 2 and 3 represent the second examples of noremestrin besides the previously reported noremestrin A, and the single crystal X-ray diffraction analysis of compound 2 provided solid evidence for the intriguing skeleton of noremestrin. Their structures were determined by extensive spectroscopic data, electronic circular dichroism calculations, and single-crystal X-ray diffraction.

Analysis of glass-based artworks is important for authentication purposes. In recent years, there have been rapid advancements and improvements in the characterization of glass objects using different analytical approaches. Surface relief grating formation in photo-responsive azo polymers under irradiation is a long-ago-found phenomenon, but all the factors governing its efficiency are still not fully recognized. Here, we report on the enormous impact of the polymer thickness on the possibility of fabrication of extremely high-amplitude surface deformations. We performed prolonged holographic recordings on the layers of the same azobenzene poly(ether imide), which had substantially different optical transmittances at the recording wavelength and revealed that the depths of the inscribed relief structures increased with the polymer thickness from a nondetectable value up to almost 2 µm, unaffected by the presence of a polymer-glass substrate interface in either sample.
