Recent advances in the digitization of manufacturing have prompted ASME and International Organization for Standardization (ISO) standards committees to reexamine the definition of datums. Any new definition of datums considered by the standards committees should cover all datum feature types used in design and support both traditional metrological methods and new digital measurement techniques. This is a challenging task that requires some careful compromise. This paper describes and analyzes various alternatives considered by the standards committees. Among them is a new mathematical definition of datums based on constrained least-squares fitting. It seems to provide the best compromise and has the potential to support advanced manufacturing that is increasingly dependent on digital technologies.

The Upper Cretaceous (Campanian) Belly River Group (BRG) of southern Alberta has a complex internal stratigraphic architecture derived from differential geometries of its component formations that resulted from regionalized tectonic influences and shifting source areas. A full understanding of BRG architecture has been compromised heretofore by a limited understanding of subsurface data in southwestern- and southeastern-most Alberta. In this study outcrop exposures throughout southern Alberta are tied to reference well logs and subsurface cross-sections allowing a more precise understanding of BRG architecture and how it relates to well-known vertebrate fossil producing areas.

This work refines surface registration methods for metrological datasets to improve the multi-method qualification accuracy of additively manufactured (AM) lattices. Datasets acquired from X-ray computed tomography and a coordinate measurement machine of an AM lattice were aligned using derived geometry datum features based on a theoretical supplemental surface definition, which has been established in recent draft standards, but has had limited examination using complex AM structures. A refined sampling registration approach for lattice geometry based on spatially-dependent subsampling is derived and shown to statistically decrease variation between measurement sources.

Next-generation sequencing (NGS)'s crucial role in supporting genetic diagnosis and personalized medicine leads to the definition of Guidelines for Diagnostic NGS by the European Society of Human Genetics. Factors of different nature producing false-positive/negative NGS data together with the paucity of internationally accepted guidelines providing specified NGS quality metrics to be followed for diagnostics purpose made the Sanger validation of NGS variants still mandatory. We reported the analysis of three cases of discrepancy between NGS and Sanger sequencing in a cohort of 218 patients.
