The choice of organism to host a genetic circuit, the chassis, is often defaulted to model organisms due to their amenability. The chassis-design space has therefore remained underexplored as an engineering variable. In this work, we explored the design space of a genetic toggle switch through variations in nine ribosome binding site compositions and three host contexts, creating 27 circuit variants.

Degradation tags, otherwise known as degrons, are portable sequences that can be used to alter protein stability. Here, we report that degron-tagged proteins compete for cellular degradation resources in engineered mammalian cells leading to coupling of the degradation rates of otherwise independently expressed proteins when constitutively targeted human degrons are adopted. We show the effect of this competition to be dependent on the context of the degrons.

In public health, risk experts often define acceptable risk targets without community input. We developed a novel method for applying behavioral microeconomics to integrate individuals' risk preferences into risk assessment. To demonstrate this methodology, we explored a risk-risk tradeoff case scenario: increased asthma risk from increased cleaning and disinfection (C&D) and increased infection risk from decreased C&D for healthcare staff.

Respiratory interventions including noninvasive ventilation, continuous positive airway pressure and high-flow nasal oxygen generated infectious aerosols may increase risk of airborne disease (SARS-CoV-2, influenza virus) transmission to healthcare workers. We developed and tested a prototype portable UV-C device to sterilize high flows of viral-contaminated air from a simulated patient source at airflow rates of up to 100 l/m. Our device consisted of a central quartz tube surrounded 6 high-output UV-C lamps, within a larger cylinder allowing recirculation past the UV-C lamps a second time before exiting the device.
