In this work, we developed a smart drug delivery system composed of poly (ethylene glycol)--poly (ε-caprolactone) (PEG-PCL)-based polymersomes (Ps) loaded with doxorubicin (DOX) and vemurafenib (VEM). To enhance targeted delivery to malignant melanoma cells, these drug-loaded nanovesicles were conjugated to the oxalate transferrin variant (oxalate Tf) and incorporated into three-dimensional chitosan hydrogels. This innovative approach represents the first application of oxalate Tf for the precision delivery of drug-loaded polymersomes within a semi-solid dosage form based on chitosan hydrogels. These resulting semi-solids exhibited a sustained release profile for both encapsulated drugs. To evaluate their potency, we compared the cytotoxicity of native Tf-Ps with oxalate Tf-Ps. Notably, the oxalate Tf-Ps demonstrated a 3-fold decrease in cell viability against melanoma cells compared to normal cells and were 1.6-fold more potent than native Tf-Ps, indicating the greater potency of this nanoformulation. These findings suggest that dual-drug delivery using an oxalate-Tf-targeting ligand significantly enhances the drug delivery efficiency of Tf-conjugated nanovesicles and offers a promising strategy to overcome the challenge of multidrug resistance in melanoma therapy.

Fruit preservation materials play an instrumental role in preventing fruit deterioration and extending shelf life. However, existing fresh-keeping materials often prove inadequate in simultaneously achieving antibacterial properties, maintaining freshness, antioxidant effects, good biocompatibility, and prolonged fruit shelf life. Therefore, we present the first preparation of a natural polysaccharide spray hydrogel (Q/O/Zn hydrogel), loaded with chlorogenic acid‑zinc nanoparticles (CA@ZnNPs), utilizing quaternary ammonium insect chitosan (QECS) and oxidized pullulan (OPUL) for the preservation of perishable fruits.

Marine and terrestrial organisms often utilise EGF/EGF-like domains in wet adhesives, yet their roles in adhesion remain unclear. Here, we investigate the Barbatia virescense byssal system and uncover an oxidation-independent, reversible, and robust adhesion mechanism where EGF/EGF-like domain tandem repetitions in adhesive proteins bind robustly to GlcNAc-based biopolymer. EGF/EGF-like-domain-containing proteins demonstrate over three-fold superior underwater adhesion to chitosan compared to the well-known strongest wet-adhesive proteins, mefp-5, and suckerin, when adhering to mica in an surface forces apparatus-based measurement.

Achieving a synergy of biocompatibility and extreme environmental adaptability with excellent mechanical property remains challenging in the development of synthetic materials. Herein, a "bottom-up" solution-interface-induced self-assembly strategy is adopted to develop a compressible, anti-fatigue, extreme environment adaptable, biocompatible, and recyclable organohydrogel composed of chitosan-lignosulfonate-gelatin by constructing noncovalent bonded conjoined network. The ethylene glycol/water solvent induced lignosulfonate nanoparticles function as bridge in chitosan/gelation network, forming multiple interfacial interactions that can effectively dissipate energy.

Mechanical metamaterials exhibit several unusual mechanical properties, such as a negative Poisson's ratio, which impart additional capabilities to materials. Recently, hydrogels have emerged as exceptional candidates for fabricating mechanical metamaterials that offer enhanced functionality and expanded applications due to their unique responsive characteristics. However, the adaptability of these metamaterials remains constrained and underutilized, as they lack integration of the hydrogels' soft and responsive characteristics with the metamaterial design.
