Bioluminescence is a powerful method for imaging , but applications at the microscale are far from routine. This is due, in part, to a lack of versatile tools for visualizing dynamic events. To address this void, we developed a new platform-Bioluminescence Resonance Energy mAKe over with a Fluorescence-Activating absorption-Shifting Tag (BREAKFAST). BREAKFAST features a bright luciferase combined with a chemogenetic tag (pFAST) for rapid color switching. In the presence of luciferin and a discrete fluorogenic ligand, signal is observed resonance energy transfer. We evaluated spectral outputs with various fluorogens and established the utility of BREAKFAST for combined fluorescence and bioluminescence imaging. Dynamic, four-color visualization was achieved with sequential ligand addition and spectral phasor analysis. We further showed selective signal quenching with a dark fluorogen. Collectively, this work establishes a new method for bioluminescence imaging at the cellular scale and sets the stage for continued probe development.

Detailed DFT studies of H and C NMR chemical shifts of hydroxy secondary oxidation products of various geometric isomers of conjugated linolenic acids methyl esters are presented. Several low energy conformers were identified for model compounds of the central dienenol OH moiety, which were found to be practically independent on the various functionals and basis sets used. This greatly facilitated the minimization process of the geometric isomers of conjugated linolenic acids methyl esters.

Polyelectrolyte complex (PEC) hydrogels provide a promising strategy to develop a class of physically cross-linked networks characterized by exceptional toughness and self-healing properties. However, the precise control of the microstructure and the enhancement of mechanical properties still pose challenges in the field of PEC hydrogels. Herein, we propose a strategy to manipulate the structure of PEC with competitively charged surfactant micelles, leveraging the spatially confined surface charge and excluded volume effects to overcome coacervation issues associated with the PEC, thus achieving a simple one-step preparation of macroscopically uniform and tough PEC hydrogels.

The early fault characteristics of rolling bearings are weak, especially in a strong noise environment, which are more difficult to extract; therefore, a method based on wavelet packet decomposition, multi-verse optimizer, and maximum correlated kurtosis deconvolution for weak fault feature extraction of rolling bearings is proposed. First, the original vibration signal is decomposed using wavelet packet decomposition, followed by proposing a signal reconstruction method combining the Pearson correlation coefficient and energy ratio to effectively remove noise from the original signal. Second, the parameters L and M of Maximum Correlated Kurtosis Deconvolution (MCKD) are optimized using the multi-verse optimizer algorithm to obtain optimal filter settings.
