The sex determination in mammals refers to the development of an initial bipotential organ, termed the bipotential gonad/genital ridge, into either a testis or an ovary at the early stages of embryonic development, under the precise regulation of transcription factors. SOX9 (SRY-box transcription factor 9) is a multifunctional transcription factor in mammalian development and plays a critical role in sex determination and subsequent male reproductive organs development. Recent studies have shown that several enhancers upstream of also play an important role in the process of sex determination. In this review, we summarize the progress on the role of and its gonadal enhancers in sex determination. This review will facilitate to understand the regulatory mechanism of sex determination of and provides a theoretical basis for the further development of animal sex manipulation technologies.

Identifying sex from an unknown dried blood spot (DBS), especially when the corpse remains undiscovered, often provides valuable evidence in forensic casework. While DNA-based sex determination is a reliable method in forensic settings, it requires expensive reagents and is time-consuming. Over the past decade, research on embryo-derived extracellular vesicles (EVs) has unveiled their critical roles in embryonic development and intercellular communication. EVs secreted by embryos are nanoscale lipid bilayer vesicles that carry bioactive cargo, including proteins, lipids, RNAs, and DNAs, reflecting the physiological state of the source cells. These vesicles facilitate paracrine and autocrine signaling, influencing key processes such as cell differentiation, embryo viability, and endometrial receptivity.

The current study is a forerunner analysis that used three-dimensional multidetector computed tomography (3D MDCT) images of the hyoid bone to examine sexual dimorphism in the Egyptian population. Global trends in marine turtle nesting numbers vary by region, influenced by environmental or anthropogenic factors. Our study investigates the potential role of past temperature fluctuations on these trends, particularly whether warmer beaches are linked to increased nesting due to higher female production (since sea turtles have temperature-dependent sex determination). We selected the loggerhead turtle (Caretta caretta) due to its wide distribution, strong philopatry and vulnerability to environmental changes.
