Hydrogels are extensively used in the biomedical field due to their highly valued properties, biocompatibility and antimicrobial activity and resistance to rheological stress. However, determining an efficient sterilization protocol that does not compromise the functional properties of hydrogels is one of the challenges researchers face when developing a material for a medical application. In this work, conventional sterilization methods (steam-, radiation- and gas sterilization) were investigated regarding the influence on the degree of swelling, mechanical performance and chemical effects on the poly 3-sulfopropyl acrylate potassium (pAESO) hydrogel, which is a promising representative for biomedical engineering applications. In summary, no significant changes in the gel properties were observed after sterilization, showing the potential of the selected hydrogel for biomedical applications.

This study investigates hydrogels based on 2-Acrylamido-2-methyl-1-propanesulfonic acid sodium salt (AMPS) copolymers, incorporating N-hydroxyethyl acrylamide (HEA) and 3-sulfopropyl acrylate potassium salt (SPA). The addition of HEA and SPA is designed to fine-tune the hydrogels' water absorption and mechanical properties, ultimately enhancing their characteristics and expanding their potential for biomedical applications. A copolymer of AMPS, 2-carboxyethyl acrylate (CEA) combined with methacrylic acid (MAA) as poly(AMPS-stat-CEA-stat-MAA, PACM), was preliminarily synthesized.

Cryogel microcarriers made of poly(ethylene glycol) diacrylate and 3-sulfopropyl acrylate have the potential to act as delivery vehicles for long-term retention of neurotrophic factors (NTFs) in the brain. In addition, they can potentially enhance stem cell-derived dopaminergic (DAergic) cell replacement strategies for Parkinson's disease (PD), by addressing the limitations of variable survival and poor differentiation of the transplanted precursors due to neurotrophic deprivation post-transplantation in the brain. In this context, to develop a proof-of-concept, the aim of this study was to determine the efficacy of glial cell line-derived NTF (GDNF)-loaded cryogel microcarriers by assessing their impact on the survival of, and reinnervation by, primary DAergic grafts after intra-striatal delivery in Parkinsonian rat brains.

Microcarriers for large-scale cell culture have a broader prospect in cell screening compared with the traditional high cost, low efficiency, and cell damaging methods. However, the equal biological affinity to cells has hindered its application.
